Abstract

Calcium reacting photoproteins are a class of self-illuminating proteins that emit light upon contact with calcium. Fluorescent Proteins are small proteins that change the color of light when excited. Fluorescent Proteins and Calcium Activated Photoproteins can be used to enhance, dazzle, amaze, startle, and otherwise entertain an audience by their direct application on to the audience, surroundings, the actors, or sprayed on settings as in the newer 4D movies. The disclosure comprises novel Coelenterazine compounds and methods of use, including a simple delivery device for the photoprotein to create unique and novel cinematic, theatrical, stage, movie and musical concert optical effects by their luminous reaction upon contact with surfaces that contain calcium. Calcium is ubiquitous in and on most surfaces and in the environment; it is this unique property of calcium that makes this a novel use of the photoproteins for entertainment. Abstract

The present invention relates to the extraordinary properties of recently discovered nanotubes. This disclosure teaches a method for using barrel proteins acting as scaffolds to guide assembly of nanotubes, and using nano-molecular molding jigs to format the nanotubes into stable arrays with the precise geometric architecture desired. This disclosure teaches nanotube technology with principles of protein folding and aggregated self-assembly. In certain embodiments, the disclosure teaches using highly modified barrel proteins to form hydrophobic and hydrophilic channels that guide the nanotubes into their centers, or other geometric patterns utilizing silicone aerogel to form nano-molecular molds, jigs, and surfaces to position nanotubes in precise geometric arrangements and arrays. This disclosure teaches new uses of barrel proteins as self-assembling molding tools to develop new nanometer scaled devices and their uses herein.

Abstract

Methods and compositions are described for assaying luciferase bioluminescence in vitro and in vivo cells. The compositions provide at least one of enhanced stability of signal or magnitude of signal by varying the composition of the buffer. One or more of the following parameters have been varied: the presence or absence of EDTA, the concentration of NaCI, the concentration of coelenterazine, the evaluation of ionic and non-ionic detergent, the amount of detergent, how the detergent has been added and the time over which the signal has been recorded.
