Abstract

Disclosed is a fabrication method for constructing low-cost, morphologically stable, highly ordered, and crystalized layered organic solar cells. The method implements self-assembled molecular monolayers as building blocks (a bottom up strategy) to fabricate a device. This approach enables the creation of a layered material with desired morphology in a controlled way. In such geometry, optoelectronic and transport properties can be controlled by metal atom inclusions into the molecular monolayers, which presents a range of options in creating photo-sensitive molecular building blocks to cover a wide range of the solar spectra from IR to visible to UV.

Abstract

The SARS-CoV-2 main protease, MPRO, is critical for its replication and is an appealing target for designing anti-SARS-CoV-2 agents. In this regard, a number of assays have been developed based on its cleavage sequence preferences to monitor its activity. These include the usage of Fluorescence Resonance Energy Transfer (FRET)-based substrates in vitro and a FlipGFP reporter, one which fluoresces after MPRO-mediated cleavage, in live cells. Here, a pair of genetically encoded, Bioluminescence Resonance Energy Transfer (BRET)-based sensors have been engineered for detecting SARS-CoV-2 MPRO proteolytic activity in living host cells. The sensors were generated by sandwiching MPRO N-terminal autocleavage sites, either AVLQSGFR (short) or KTSAVLQSGFRKME (long), in between the mNeonGreen and nanoLuc proteins. Co-expression of the sensor with the MPRO in live cells resulted in its cleavage in a dose-dependent manner while mutation of the critical C145 residue (C145A) in MPRO completely abrogated the sensor cleavage. A temporal activity of MPRO in live cells and its inhibition was shown using the well-characterized pharmacological agent GC376. The sensor developed here finds direct utility in studies related to drug discovery targeting the SARS-CoV-2 MPRO and functional genomics application to determine the effect of sequence variation in MPRO Importantly, the BRET-based sensors displayed increased sensitivities and specificities as compared to the recently developed FlipGFP-based MPRO sensor. Additionally, the sensors recapitulated the inhibition of MPRO by the well-characterized pharmacological agent GC376. Further, in vitro assays with the BRET-based MPRO sensors revealed a molecular crowding-mediated increase in the rate of MPRO activity and a decrease in the inhibitory potential of GC376. The sensor developed here finds direct utility in studies related to drug discovery targeting the SARS-CoV-2 MPRO and functional genomics application to determine the effect of sequence variation in MPRO.

Abstract

A method for synthesis of fractal metallic nanostructure is provided. The method includes applying electron-beam irradiation directly on a metal-containing carbon nanosheet. Moreover, the Disclosed Invention relates to a novel method for the direct synthesis and control of fractal growth of metallic nanostructures on hybrid self-assembled metal/carbon nanosheets using electron-beam irradiation. In particular, the nanosheet is the source or the metallic precursor for the fractal nanostructure as well as the substrate on which the fractal formation takes place. In addition, the irradiation-induced interactions between the electron-beam and the carbon-based nanosheet enables the patterning and carving of the nanosheet with different designs of various complexities to eventually control the path and route of the irradiation induced nucleation and growth of the metallic fractal morphology.

Abstract

The present disclosure provides a therapeutic method comprised of targeting the tumor-associated antigen, Lactate Dehydrogenase C (LDHC), in LDHC-expressing cancers or tumors. This novel approach offers a highly specific cancer treatment with little or no off-target effects. Targeting LDHC with the use of silencing molecules increases tumor cell death by inducing excess DNA damage, and more importantly, can render breast tumor cells more responsive to non-specific, DNA damaging drugs and DNA damage repair inhibitors, such as platinum-based alkylating agents (e.g. cisplatin) and Poly ADP-Ribose Polymerase (PARP) inhibitors (e.g. olaparib). Targeting LDHC in tumor cells has the potential to specifically sensitize tumor cells to these drugs and/or inhibitors, leading to enhanced tumor cell killing and potentially minimizing side effects.

Abstract

A high-oxidation and NOx-free synthesis of graphene oxide (GO) from natural graphite using the modified Hummers' method is described. The amine-functionalized GO using dodecylamine (DDA) is used as a filler for membranes for the first time. Antifouling and antibacterial properties of UF membranes are achieved using amine functionalization of GO. A process of incorporating raw GO and dodecylamine-functionalized GO (GO-DDA) in polysulfone (PSF) via phase inversion technique is disclosed.
