Abstract

A method for producing an HSL protein with increased catalytic activity to oxidize a 4-HPPD inhibitor in a 2-oxoglutarate-dependent manner is disclosed. A method for producing a plant with increased resistance to a 4-HPPD inhibitor using the method for producing the HSL protein is also disclosed. It has been revealed that, by mutating position 140 to a basic amino acid in an HSL protein, the catalytic activity of the protein to oxidize a 4-HPPD inhibitor in a 2-oxoglutarate-dependent manner can be increased, and an activity of the protein to decompose the inhibitor can be increased.

Abstract

By a QTL analysis and so forth using 4-HPPD inhibitor-susceptible rice and 4-HPPD inhibitor-resistant rice, a hypothetical gene (HIS1 gene) of an iron/ascorbate-dependent oxidoreductase gene located on a short arm of chromosome 2 of rice has been identified as a 4-HPPD inhibitor-resistance gene. Further, it has also been revealed that a homologous gene (HSL1 gene) of the HIS1 gene is located on chromosome 6 of rice. Abstract

The present invention pertains to: a tobacco axillary bud growth inhibitor which comprises both chlorthal-dimethyl and an aliphatic alcohol selected from among decyl alcohol, 2-ethylhexanol and geraniol; and a method for inhibiting tobacco axillary bud growth, said method including applying said tobacco axillary bud growth inhibitor. The tobacco axillary bud growth inhibitor exhibits excellent persistency of drug efficacy even at a low concentration, does not induce chemical injury or disease, and can improve labor productivity.

As the active ingredient compound, chlorthal-dimethyl, butralin, pendimethalin or decyl alcohol is preferred. As the surfactant, a silicone-based surfactant or a fluorine-based surfactant is preferred. The contact-type inhibitor for tobacco axillary bud growth according to the present invention exhibits a high axillary bud growth inhibition effect at a low concentration, has excellent sustained drug efficacy, induces neither chemical injury nor disease, and can contribute to the improvement in labor productivity.

A 4-HPPD inhibitor-resistant gene is identified as a gene (HIS1 gene) which is estimated as an iron-/ascorbic acid-dependent oxidoreductase gene located on the short arm of chromosome-2 of a rice plant, by carrying out QTL analysis using a 4-HPPD inhibitor-sensitive rice plant and a 4-HPPD inhibitor-resistant rice plant or the like. It is also found that a homologous gene (HSL1 gene) to the HIS1 gene is located on chromosome-6 of a rice plant. It is found that a plant having improved resistivity or sensitivity to a 4-HPPD inhibitor can be produced with high efficiency and the resistivity or sensitivity of a plant to a 4-HPPD inhibitor can be determined with high efficiency using the aforementioned genes.

A nucleic acid fraction or a virus fraction in a sample is extracted and introduced into a target host cell by a method of introducing a nucleic acid into a cell noninfectiously and directly. By introducing the nucleic acid fraction or the like into the target host cell directly like this, the virus can be replicated in the host cell without the influence of virus infectivity to the host cell and a mature virus can be isolated. As the introduction method, for example, a calcium phosphate method, a polyethyleneglycol method, a lipofection method, an electroporation method, an ultrasound transfection method, an introduction method utilizing a gene gun or the like can be used.
