Abstract

Provided are a cutinase mutant and the use thereof in the efficient degradation of PET. The amino acid sequence of the cutinase mutant is as shown in SEQ ID NO.2, where phenylalanine at position 239 of the amino acid sequence of the wild-type cutinase as shown in SEQ ID NO.1 is replaced with alanine. The cutinase mutant constructed in the present invention is expressed in Escherichia coli. Compared with the wild-type enzyme, the activity of the mutant enzyme shows increased stability, and improved catalytic efficiency, such that the yield of the degradation product of PET plastics (polyethylene terephthalate, PET) is increased by 42.6 times. The modified cutinase mutant improves the PET degradation efficiency.

Abstract

A novel disturbance-free marine sediment sampling tool, comprising an outer cylinder (2), an inner cylinder (1), a turnover lever assembly (3) and a petal assembly (4). When the sampling tool is moving downwards, the petal assembly (4) is clamped by utilizing an upward acting force of water flow and sediments on the turnover lever assembly (3), so as to keep the petal assembly (4) in an open state during the whole sampling process. When the sampling is completed and the sampling tool starts to be lifted upwards, the petal assembly (4) is pried by utilizing a downward acting force of the sediments on the turnover lever assembly (3), so as to close the petal assembly (4). The sampling tool utilizes the natural resistance of water flow and sediments, etc., and by means of the linkage of the turnover lever assembly (3) and the petal assembly (4), the problem of disturbance and resistance generated by the petal assembly (4) on sediment sampling is effectively solved, and the sampling efficiency is improved while leakage of the sediment sample is effectively blocked.

Abstract

A method and apparatus for inverting a crustal structure of a passive continental margin on the basis of subsidence in a stratigraphic record. The method comprises seven steps. In comparison with the prior art, without the need to additionally provide gravity or refraction seismic data and simply by means of reflection information of a multichannel seism to a stratum, a sedimentary basement and the Moho, the crustal average density, speed and thickness can be quickly and efficiently calculated within a large range to establish a crustal attribute and a depth structure. An effective means is provided for obtaining space variation information of a crustal structure of a passive continental margin that lacks data, and an important basis is thus provided for understanding the formation, basin formation and reservoir formation mechanisms of the passive continental margin.

A deep-sea in-situ long-term experimental platform having a sediment sampling function, which relates to the field of deep sea apparatuses. The platform enables, by means of a multi-stage release mechanism, buoyancy to match gravity to complete an entire sampling recovery process of sediment, which only requires small power units to be sequentially triggered to complete intubation and sampling without a powerful power source to provide kinetic energy for vertical movement. Disclosed in the present invention is an intelligent solution injection device for a deep-sea sediment microorganism culture experiment. The intelligent solution injection device is mainly composed of an insertion device, a controllable slow-release solution injection system, an interstitial water displacement and monitoring system, and a deep-sea peristaltic pump. The insertion device can insert a solution injection needle tube and an interstitial water collection tube into a designated sediment position by means of gravity or other driving devices; the controllable slow-release solution injection system is used for receiving an instruction to inject a certain amount of culture solution into sediment through the solution injection needle tube; and the interstitial water displacement and monitoring system is used for collecting sediment interstitial water and monitoring environmental parameters to develop a solution injection policy. The present invention can simulate a diffusion process of matter seeping out of a deep-sea cold seep, and can provide a powerful tool for researchers to conduct long-term in-situ experiments of deep-sea sediment microorganisms.

Disclosed are a Litopenaeus vannamei vibrio-resistance related EST-STR marker, and specific primers thereof and a detection method therefor. A constructed transcriptome library is utilized to develop and obtain six Litopenaeus vannamei EST-STR markers with high polymorphism, numbered Lv-Vp001, Lv-Vp011, Lv-Vp012, Lv-Vp023, Lv-Vp029 and Lv-Vp041, respectively, and specific primers thereof are respectively designed. An environment-friendly composite basalt fiber reef base grid suitable for coral reef substrate restoration and a restoration method, comprising a square grid woven by gel-free basalt fibers. Pure aluminum wires are embedded in each bundle of gel-free basalt fiber; sleeves made of basalt fiber cloth are fixedly connected to four edges of the grid, and a sleeve made of basalt fiber cloth is also provided in the middle of the grid; fixed connection holes are formed in four corners of the grid. A stable measurement installation support for magnetotellurics magnetobars, comprising insertion rods. The insertion rods are inserted into dug holes or into the ground so as to fix the position of a bottom panel. Abstract

Provided is an artificial ecological reef made of a composite concrete material for the ecological restoration of coral reefs. The artificial ecological reef comprises a concrete reef body, wherein iron fibers are at least distributed on a surface of the concrete reef body. According to the artificial ecological reef of the composite concrete material, a surface is provided with the uniformly distributed high-density and high-carbon cast iron fibers, wherein same are increased by 25% to 30% compared with that in a common concrete reef. The concrete reef has rich three-dimensional structures and may provide more spaces for coral larvae to adhere thereto, and this may improve the adhesion rate of the coral larvae. Moreover, since the carbon content of the high-carbon cast iron fibers is high, iron molecules and carbon molecules in high-iron fibers are likely to form the metal of a galvanic cell, such that an oxidation reaction takes place actively, and the resulting biochemical reaction can promote the calcification growth of hermatypic corals.

An environmentally friendly artificial mimic coral reef body widely applied to the construction of reefs at a coral ecological point and to the reconstruction of three-dimensional structures in a reef area in coral reef ecosystem restoration. For engineering repair of coral reef ecosystems of open sea island reefs, lagoons and offshore waters, an artificial mimic reef body highly similar to natural coral reefs in form and function is constructed through simulation design. This mimic reef body is built using a unique environmentally friendly raw material with an irregular form and has many structural features such as large internal gaps, a rough reef body surface, a large surface area and high spatial heterogeneity, and has advantages such as high hardness, being non-polluting and having a high biological attachment rate. This mimic reef body can provide a growth substrate for critical reef-creating organisms for the coral reef ecosystem such as coral, giant clam and reef-creating seaweed and provide a growth, habitat and reproduction space for other reef protection biological groups.

Abstract

Disclosed are an SNP marker related to the low-salt tolerance characteristic of Litopenaeus vannamei, an amplification primer therefor, and an application thereof. A primer is designed on the basis of the mRNA sequence (GenBank: KF765670.1) of the Na,K-ATPase α subunit of Litopenaeus vannamei obtained from NCBI, and the genomic DNA of low-salt tolerant and low-salt intolerant Litopenaeus vannamei are used as templates respectively for PCR amplification to obtain PCR products with a length of 533 bp respectively. The sequencing results are compared and analyzed. A total of 11 SNP sites are detected in this sequence. An SNP marker related to low-salt tolerance is identified by correlative analysis of the genotypes of these templates at each SNP site.

Abstract

A preparation method for a mesoporous iron disulfide/silica microsphere composite solid-phase iron source, and an application thereof, using a sol-gel process on a natural pyrite surface to form a mesoporous SiO2 microsphere structure and prepare a mesoporous FeS2/SiO2 microsphere composite solid-phase iron source, and combining same with H2O2 to form a new Fenton oxidation system, for use in removing ciprofloxacin from bodies of water. The mesoporous FeS2/SiO2 microsphere compound solid-phase iron source can both effectively control an iron release rate of a mineral, lowering loss of a mineral iron source itself and operating costs caused by high use of H2O2, and also avoid problems of secondary environmental pollution caused by high release of Fe3+. A system and method for testing an adiabatic stress derivative of temperature of underwater rock. By means of the method, without the use of an electrical heating heat source and a booster pump, instantaneous loading of a rock sample can be achieved by quickly opening a bleeder valve, and thermophysical properties of the rock sample under a high pressure condition are obtained by means of an established finite element numerical inversion model.

The system is simple and reasonable in structure, and can truly realize the adiabatic pressurization or depressurization of the rock samples, so that changes in the confining pressure of the pressure-resistant tanks (A, B) and the temperature of the rock samples are monitored in real time, so as to accurately acquire the temperature response coefficient of rock adiabatic stress changes.

Abstract

A breeding method for obtaining heterosis for a lined seahorse (Hippocampus erectus) comprises the following steps: S1 selecting parents from lined seahorse groups having greatly different genetic backgrounds; S2 intensively nurturing the parents before birth; S3 pairing parents from geographically different lined seahorse groups for breeding according to a complete diallel cross; S4 attentively caring for pregnant seahorses; S5 respectively collecting all hatched larval fishes (filial generations) of each breeding pair within one week; S6 nurturing the filial generations; and S7 comparing survival rates and growth rates of the filial generations. By hybridizing lined seahorses from geographically different groups to obtain hybrids, and effectively using the heterosis, compared with non-hybridized second generations, the survival rates and growth rates of the filial generations are increased significantly, thereby increasing genetic diversity of lined seahorses and facilitating selection of advantageous breeds thereof.

An in-situ and on-line acoustic measuring system for natural gas flux at the hydrocarbon seeps at a seafloor includes a seepage tent and a flow measuring channel. A calculation method for a seafloor in-situ thermal conductivity and a detection device using the same, which relate to the technical field of seafloor in-situ heat flow detection. A gesture detection module detects a gesture and the movement condition of a probe at a seafloor, and then a main control module controls a heat pulse control module to excite a heat pulse, so as to perform electric heating on a heating wire of the probe at the front end of a measurement unit; the main control module controls a temperature collection module to collect temperature data of a submarine sediment, and then the temperature data is output to a PC machine; and a finite element numerical model is established by means of the PC machine, and parameter inversion is performed by using a grid encryption search method. The gesture detection module determines seafloor arrival conditions and performs automatic heat pulse control; an in-situ environment temperature and the in-situ thermal conductivity of a submarine sediment are acquired, so that the offshore operation safety and seafloor detection efficiency can be greatly improved; and by establishing an in-situ thermal conductivity numerical inversion method, the limitation of an analytical solution of a simplified model depended by the in-situ thermal conductivity solution is eliminated.

A breeding method for a rapid growth strain of Hippocampus kelloggi. The breeding method comprises: (1) selecting a high-quality Hippocampus kelloggi parent as a breeding base population; (2) intensively culturing before reproduction; (3), performing paired reproduction and culturing bred fry; (4), setting different selection fractions at three critical periods and gradually breeding; (5), pairing selected adult fishes; during pairing, eliminating individuals with poor activity, body injury or pouch or gonadal dysgenesis; afterwards, controlling the Hippocampus kelloggi of each generation to breed at the selection fraction of 10%; and (6), repeating step (2) to step (5) for at least four times to obtain the high-quality Hippocampus kelloggi strain with greatly-improved body weight and body length and stable growth. By means of the breeding method, high-quality and high-yield new strains can be bred, and the problems that the fry of the current Hippocampus kelloggiare are slow in growth, are irregular in size and the like are solved from the germplasm.

A self-floating seafloor heat flow long-term observation station comprises a recycling unit (1), a discarding unit (2) and a cable cutting mechanism (3). The recycling unit (1) is provided with a recycling support, the recycling support is internally provided with two acoustic release transponders (13), the bottoms of the acoustic release transponders (13) are provided with hooks capable of being closed (131), and the recycling support is provided with floating balls (14) in a carrying mode. The discarding unit (2) is provided with a discarding support, and a heat flow probe (24) is fixedly connected below the discarding support. The recycling unit (1) and the discarding unit (2) are fixed together through steel wire ropes (4) of which the two ends are connected with the hooks capable of being closed and arranged at the bottoms of the acoustic release transponders (13). The cable cutting mechanism (3) is fixed to the bottom of the recycling support of the recycling unit (1) and is connected with the steel wire ropes (4) through the movable hooks. Cables (0) start off from the discarding unit (2), then enter the cable cutting mechanism (3), and then penetrate out of the cable cutting mechanism (3) and are connected with the floating balls (14) of the recycling unit (1). Abstract

An impact pin-type cable severing mechanism consisting of a blade casing, a cable press plate, an ejection control casing, and a movable hook. Provided within the blade casing are a blade and a powerful compression spring below the blade. The ejection control casing is connected with the movable hook. A trigger piece and a blade latch are provided within the ejection control casing. In the impact pin-type cable severing mechanism, when the hook is taut, the blade latch within the ejection control casing is utilized to lock the blade, thus allowing the compression spring to remain in a compressed state, and when the hook is slack, the trigger piece is impacted by a rebound force to drive the blade latch into separating from the blade, and the compression spring is released to eject the blade to the cable press plate to sever a cable. An in-situ ultrasonic measurement system for seepage flow of seafloor cold spring natural gas. The system comprises a seepage tent cover (2) and a flow measurement channel (1). The flow measurement channel (1) comprises a lower bubble breaking channel (11), an ultrasonic sensor measurement channel (12) and an upper bubble breaking channel (13) that are arranged from bottom to top in sequence and are communicated with each other. The lower bubble breaking channel (11) is communicated with the seepage tent cover (2) provided with bubble breaking grids (3). A lower bubble breaking device (4) and an upper bubble breaking device (5) that are arranged in the bubble rising direction are respectively mounted in the lower bubble breaking channel (11) and the upper bubble breaking channel (13). One side of the ultrasonic sensor measurement channel (12) is fixedly connected to an acoustic wave branching unit (7), and the other side is fixedly connected to a flat receiving transducer (8) used for receiving transmitting acoustic waves generated by the acoustic wave branching unit (7). An acoustic wave probe (9) also used for receiving the transmitting acoustic waves generated by the acoustic wave branching unit (7) is arranged in the ultrasonic sensor measurement channel (12). The ultrasonic measurement system can be used for performing long-term on-line measurement on in-situ seepage flow of seafloor cold spring natural gas in the environments of different water depths.

Disclosed is a bee venom composition with a sun-proof effect. The bee venom composition comprises a sun-proof cosmetic substrate component, and further comprises bee venom. Abstract

Disclosed in the present invention are three cycloheptapeptide compounds; the compounds are isolated from the fermentation products of a marine fungus Acremonium persicinum SCSIO 115, of which the structure is shown in formula (I), wherein R1=H, R2=H in compound 1, R1=OH, R2=H in compound 2 and R1=OH, R2=Me in compound 3. The compounds have an obviously inhibiting effect on the glioma cell strain (SF268), the breast cancer cell strain (MCF-7) and the human large cell non-cancer cell strain (NCI-H460) so can be used for the preparation of anti-tumour drugs and the treatment of tumours.

Disclosed are a Pseudonocardia sp. and a method for preparing deoxynyboquinone by the same. Pseudonocardia sp. SCSIO 01299 was deposited on July 18, 2011, at the China Center for Type Culture Collection (CCTCC), address: Wuhan University, Wuhan, China, with the deposit number CCTCC NO: M 2011255. Pseudonocardia sp. SCSIO 01299 is able to produce an antibiotic deoxynyboquinone, so Pseudonocardia sp. SCSIO 01299 can be used to prepare deoxynyboquinone, thereby providing a new way for production of the antibiotic deoxynyboquinone having the anti-tumor activity.

Abstract

A Bacillus barbaricus strain SCSIO 02429(CCTCC NO: M2010213) derived from sea is provided. Also provided is a method for preparing small squid peptide, which comprises adding Bacillus barbaricus strain SCSIO 02429 to fermentation medium inducing enzyme production to ferment and obtain crude enzyme solution used to hydrolyze, then crushing squid livers into pulp, adding the crude enzyme solution into the pulp to enzymatically digest it and obtain crude protein hydrolysate the glycosyl side chains of which are destroyed, adding bromelain to enzymatically digest, keeping the hydrolystae obtained on standing to separate crude fat from water layer, removing fat layer and drying the remains to obtain small squid peptides. The yield rate of small squid peptides by the method of the present application is 40-46%, and the yield rate of amino acids is 16-22%. The small squid peptides can be used to decrease mortality of young maricultural animals and improve growth rate. The small squid peptides also can be used to replace the traditional protein sources in feed such as fish meal, and can be used as functional protein sources and additives in maricultural formulated feed.

Abstract

Provided are a peptide extracted from squid viscera and the preparation method thereof, characterized in that the peptide comprises the amino acids fragment -ILGGSDPKHYTG-, the relative molecular mass of the peptide is in the range of 1400-5800, and the peptide is obtained by the following steps: enzymolyzing the viscera of Todarodes pacificus, filtrating with an ultrafiltration membrane with molecular mass cutoff of 18000 Da, isolating and purifying with gel chromatography, thus obtaining a peptide comprising the amino acids fragment -ILGGSDPKHYTG-, then adding potassium sorbate, stirring uniformly and drying. Also provided is a composition consisting of said peptide 6-8%, soybean meal powder 50-60%, fish powder 3-6%, corn meal powder 27-33% and starch for granulation 2-5%. The composition may be used as protein source of feed for marine aquaculture, especially protein source of feed for marine aquatic fish.
