A cell centrifugation consumable and a method of using the cell centrifugation consumable is disclosed. Abstract

The application relates to biological components, methods, systems, and kits for modulating immune responses. The disclosed biological components include genetically modified cells comprising an inserted exogenous sequence in a major histocompatibility complex (MHC)-associated gene. The inserted exogenous sequence encodes a peptide and the genetically modified cells express a fusion protein comprising the peptide and at least a portion of the polypeptide encoded by the MHC-associated gene to form a modified MHC complex. A cell centrifugation consumable and a method of using the cell centrifugation consumable is disclosed. The cell centrifugation consumable includes an inner fluid chamber configured to hold cells during centrifugation and a decanting space. The inner fluid chamber includes a supernatant fluid port that permits drawing off of supernatant from the inner fluid chamber, and a decanting port that permits a portion of the supernatant to flow into a decanting space when the cell centrifugation is tipped approximately 90°. Together, the supernatant fluid port and the decanting port allow the cell centrifugation consumable to remove the supernatant without knowing the volume of the supernatant. The cell centrifugation consumable further includes a collection chamber fluid port configured to flow additional fluid into to the cell centrifugation consumable after centrifugation, wherein the additional fluid resuspends the cell pellet.

Abstract

Various embodiments of the disclosure provide a cell processing system including: a liquid patch panel (LPP) including a flow coupler and a first flow path at least partially defined by the flow coupler. The flow coupler may include: a lumen; a hollow tube (e.g. a needle or cannula) positioned within the lumen; and a first barrier (e.g. a septum such as an engageable septum) that isolates the lumen from the ambient environment. The cell processing system may further include a container (e.g. a consumable) including a second barrier (e.g. a septum) that seals an internal volume of the container from the ambient environment. The flow coupler may be configured to move the first barrier into contact with the second barrier of the container while the hollow tube is positioned within the lumen and to move the hollow tube towards the first barrier until the hollow tube opens the first barrier, opens the second barrier, and enters into the internal volume of the container. In various embodiments of the cell processing system, the hollow tube may be presterilized.

Abstract

The present invention provides an automated modular system and method for production of biopolymers including DNA and RNA. The system and method automates the complete production process for biopolymers. Modular equipment is provided for performing production steps with the individual modules arrange in a linear array. Each module includes a control system and can be rack mounted. One side of the array of modules provides connections for power, gas, vacuum and reagents and is accessible to technicians. On the other side of the array of modules a robotic transport system is provided for transporting materials between module interfaces.
