An object is to provide a plant genome editing technique with higher genome editing efficiency. This object is achieved by a polynucleotide comprising a site-specific nuclease expression cassette and a sequence that binds to a nuclear translocation factor and/or an intranuclear structure, the site-specific nuclease expression cassette comprising (b) a site-specific nuclease coding sequence and (c) a 3′UTR comprising a terminator.

Abstract
Provided is a plant transformation technique that is more efficient and widely usable even for species in which seed gathering is difficult. (c) This method for producing a plant introduced with a polynucleotide includes introducing the polynucleotide into a callus derived from a stem apical tissue of the plant to obtain a callus introduced with the polynucleotide.
The present invention addresses the problem of providing plant genome editing technology with higher genome editing efficiency. The problem addressed by the present invention is solved by a polynucleotide including a site-specific nuclease expression cassette and a sequence that binds to a nuclear translocation factor and/or an intranuclear structure, wherein the site-specific nuclease expression cassette includes (b) a site-specific nuclease coding sequence and (c) a 3'UTR including a terminator.
An object is to provide a plant genome editing technique with higher genome editing efficiency. This object is achieved by a polynucleotide comprising a site-specific nuclease expression cassette and a sequence that binds to a nuclear translocation factor and/or an intranuclear structure, the site-specific nuclease expression cassette comprising (b) a site-specific nuclease coding sequence and (c) a 3?UTR comprising a terminator.
Provided is a grafting device that can improve productivity while suppressing variations in the quality of grafted seedlings. The present disclosure provides a grafting device for performing grafting using a grafting member including: a support member having an abutting portion with which the stem of a grafting plant abuts; and a fixed member having a pressing part for pressing the stem against the abutting portion. The grafting device comprises: a gripping part that grips the grafting plant and arranges the grafting plant on the support member; an attachment part that attaches the fixed member to the support member so that the stem of the grafting plant is pressed against the abutting portion; a cutting part that obtains a scion and a rootstock by cutting the grafting plant and divides the support member into a scion unit that holds the scion and a rootstock unit that holds a rootstock; and a joint part that joins the scion and the rootstock by attaching the rootstock unit to the scion unit.
The problem addressed by the present invention is to provide a simpler, more efficient plant genome editing technique in which gene recombination is not essential. The problem is solved by a polynucleotide including a split-Cas protein coding sequence and a transitive plant virus vector sequence.
Provided is a member for grafting use, which enables the improvement in accuracy of grafting and the productivity of a graft seedling. The present disclosure is a member for grafting use, which is provided with: a graft unit which has a first contact part that comes in contact with a stem of a plant at a first position; a rootstock unit which is attachable and detachable to and from the graft unit and has a second contact part that comes in contact with the stem of the plant at a second position that is nearer to a root of the plant than the first position; a first fixing tool which is fixable to the graft unit and has a first pressing part at which the stem of the plant can be pressed against the first contact part; and a second fixing tool which is fixable to the rootstock unit and has a second pressing part at which the stem of the plant can be pressed against the second contact part. A space in which the stem of the plant is to be exposed is provided between the first fixing tool fixed to the graft unit and the second fixing tool fixed to the rootstock unit.
