Disclosed herein are methods of making an implant material, the method involving harvesting allograft cancellous bone containing bone marrow from a donor within 24 hours of death; incubating the harvested allograft cancellous bone in an ultra-low freezer within 144 hours of death for at least 5 days; and then selectively lysing cells in the cancellous bone. This results in cancellous bone that is enriched for cells resistant to lysing, such as mesenchymal cells as well as a lysate containing factors (e.g. growth factors) released from the lysed cells. This lystate can be administered to cortical bone, e.g. demineralized cortical bone, with hydrochloric acid (HCI) or glycolytic acid so that these factors attach to the demineralized bone.

INCREASING VIABILITY OF CELLS WITHIN A FROZEN CELLULAR IMPLANT
The cryopreservation of cells from human tissue has challenges from both logistical timing as well as maintenance of cell viability. Therefore, needed are methods to increase cell viability in cellular tissue implants that allow for greater flexibility in timing for processing the implant. Therefore, disclosed herein is a method for increasing viability of cells within a frozen cellular implant, that involves soaking the cellular implant in a first cryoprotectant solution for at least 1 minute, soaking the cellular implant in a second cryoprotectant solution for at least 1 minute, and freezing the cellular implant. Also disclosed is a method for sterilizing a cellular implant comprised of collagen while maintaining cell viability that involves providing a cellular implant, adding a cryoprotectant to the cellular implant, freezing and maintaining a frozen temperature, and irradiating the cellular implant with radiation.

The harvested bone material is exposed to a lysing agent, the lysing agent configured to release growth factors and bioactive materials from cellular material of the harvested bone material.
