Abstract

The subject invention provides a novel fluorescence-based, T5 exonuclease-amplified DNA cleavage assay for gyrase poisoning inhibitor discovery. According to the assay, multiple gyrase molecules can simultaneously bind to a plasmid DNA molecule to form multiple gyrase-DNA cleavage complexes on the same plasmid. These gyrase-DNA cleavage complexes, greatly stabilized by a gyrase poisoning inhibitor, and can be trapped by a detergent such as sarkosyl. Digestion of gyrase by proteinase K results in the production of small DNA fragments, which can be digested by T5 exonuclease. This fluorescence-based DNA cleavage HTS assay is also suitable for screening large compound libraries to identify inhibitors against DNA topoisomerases, including human DNA topoisomerase IIα.

Abstract

The subject invention is concerned with systems and methods advantageously applying finite element analysis to establish design rules for a highly sensitive piezo-resistive pressure sensor with an output that is high enough to be detectable by simple and inexpensive circuits and therefore ensure wearability. Four frequently reported micro-feature shapes in micro-patterned piezo-resistive sensors are provided, where the micro-dome and micro-pyramid yield the highest sensitivity. Investigations of different conductivity values of micro-patterned elastomers show that coating the elastomer with a conductive material (e.g., a metallic coating) leads to higher current response when compared to composited conductive elastomers. Advantageous geometric parameters and spatial configurations of micro-pyramid design of piezo-resistive sensors are provided. Results show that an enhanced sensitivity and higher current output can be achieved by a lower spatial density configuration of three micro-features per millimeter length, a smaller feature size of around 100 μm, and a 60-50 degrees pyramid angle.

Abstract

Systems and methods for evaluating historical trends in real estate prices are provided. Systems and methods provide an understanding of historical price trends and assist in a property evaluation or appraisal, as well as allowing for an analysis of comparables in estimating a reasonable offer for a property on the market. Given a timespan of interest, a locale, and a category of properties of interest, an objective historical trend in values can be computed by first evaluating the ratios between the realized prices of transactions and objective governmental assessment of the properties at some fixed point of time. Then, for each period the ratios of all transactions in that period can be averaged, followed by comparing said averages between different time periods.

Abstract

Systems and methods for the control and operation of a grid-connected converter with an energy storage system are provided. The system can include a small microgrid comprising an AC grid that is feeding a DC load through a converter. The converter is connected to the AC grid through an R-L filter. The classical linear controllers have limitations due to their slow transient performance and low robustness against parameter variations and load disturbances. In certain embodiments, the transient and steady state responses of the provided artificial intelligence based Robust Artificial Neural Network Tracking Control (RANNTC) of three-phase grid-connected power converters has been shown to be more enhanced in terms of overshoot (24% lower), settling time (85% reduced), and total harmonic distortion (THD) (55% lower).

Abstract

Systems and methods for wireless simultaneous transmit and receive (STAR) using multi-input multi-output (MIMO) radiofrequency (RF) front-ends are provided. A MIMO front-end can be based on the use of a symmetrical pair of arrays, in which one is used for communication while the other is used inside an RF-shielded chamber to act as a reference for subtracting self-interference between transmitters and receivers. Abstract

Provided is a method of repairing structures on the Lunar surface using Lunar regolith containing metallic coatings. An ultra-portable and low-pressure cold spray system is utilized to consolidate coatings. This system can be easily transported to space, reducing the payload. Lunar regolith is added to the metal powder before cold spray deposition. Lunar regolith increases the tampering effect during cold spray deposition, thereby causing severe plastic deformation of the metal powders and providing a coating with high quality and minimum porosity. The presence of Lunar regolith increases the hardness of the coating and improves wear properties. Coatings on a large area can be made rapidly without much surface preparation. These coatings can be rapidly applied to various Lunar structures to repair, reinforce, and restore (RRR) functionality and performance in the Lunar environment.

Abstract

Systems and methods are provided for combined inductive power transfer (IPT) and capacitive power transfer (CPT) to reduce power pulsation in dynamic charging of electric vehicles. Aluminum plates provided between transmitter coils inhibit output power drop, the power pulsation can be reduced, and a constant power profile is provided by operation of CPT system when the receiver is between the IPT coils. As the aluminum plates cover the space between coils, the IPT coils can be separated away from each other by a longer distance compared to conventional segmented coils; therefore, a lesser copper can be used which enhances the cost effectiveness of the provided system. As the respective IPT and CPT system are used alternatively, thus the system's output power can be constant, nearly constant, or effectively constant.

Abstract

A system, apparatus, device and/or method for electrical label-free detection of single molecule via a glass or quartz nanopipette biosensor, one type of solid state nanopore biosensor based on resistive pulse measurements, are provided. The method of the subject invention effectively slows down the motion of biomolecules and concentrates them at the nanopipette tip. By driving biomolecules to come out of the nanopipette barrel and move in the fibrin hydrogel, unprecedented sensitivity using the typical size nanopipettes can be achieved. Small molecules such as doxorubicin, ATP, GTP, and short peptides can be easily detected with very high event rate, which greatly reduces the fabrication and measurement difficulties and open opportunities for various new applications for nanopipette biosensors.

Abstract

A trap accessory for attachment to a marine trap that facilitates release of by-catch. A buoyant recovery buoy is operably attached to a trap frame secured to the marine trap. Components for operably attaching the recovery buoy to the trap frame include a latch and attachment components, of which at least one of these components is a Degradable Metal (DM). The DM maintains the integrity of the trap and trap accessory for a pre-determined time, typically 1 year or less. When one or more of the DM components are sufficiently degraded, the recovery buoy is released to open a slot through which animals can escape. A buoy line connected to the trap frame maintains connection between the marine trap and the recovery buoy, which assists in recovery of the marine trap.

The subject invention provides the synthesis and use of guanidino acids and/or poly guanidino acids not only as vehicles for drug delivery but as toolbox for drug discovery. The peptidomimetic of the subject invention comprises oligo(guanidino acid)s or poly(guanidino acid)s with guanidines as peptide bond surrogates. The incorporation of the guanidine as amide bond surrogates offers significant differences in polarity, hydrogen bonding capability, and acid-base character.

Abstract

Drosophila hsp70 fusion promoter that can be used for inducible co-expression of gRNA and Cas9 in HIV-infected cells to target cellular cofactors such as Cyclin T1. A single introduction of such embodiment leads to sustained suppression of HIV replication in stringent, chronically infected HeLa-CD4 cell lines as well as in T-cell lines. In another embodiment, the invention further relates to enhancement of HIV suppression by incorporating cis-acting ribozymes immediately upstream of the gRNA in the inducible CRISPR system construct. The inducible fusion promoter is adaptable for other tissue- or cell-type specific expression of the inducible CRISPR system.

Abstract

Gradient refractive index lenses (GRI-Ls) and methods of fabricating the same are provided. GRI-Ls can be fabricated by stereolithography (SLA) and/or photo-assisted, thermal-assisted, and/or other laser-based curing from at least two precursors with a preset refractive index gradation along the planar axis. These lenses are self-focusing lenses and may be convergent or divergent for decreasing and increasing refractive indices from the center, respectively. Rather than a gradation in lens thickness from the center, the GRI-Ls can have a gradation of composition from the center.
