Abstract

The invention provides a method for predicting whether a binding peptide, which binds to a target peptide presented by a Major Histocompatibility Complex (MHC) and is for administration to a subject, has the potential to cross react with another peptide in the subject in vivo. The method comprises the steps of identifying at least one binding motif in the target peptide to which the binding peptide binds; and searching for peptides that are present in the subject that comprise the at least one binding motif and that are not the target peptide. The presence of one or more such peptides indicates that the binding peptide has the potential to cross react in vivo.

Abstract

The invention relates to screening methods. In particular, it relates to a method of identifying a functional TCR from a library of particles, which displays a plurality of different T cell receptors (TCRs). The method may comprise a) exposing the library of particles to a target antigen to identify TCRs that bind to the target antigen, b) transfecting a plurality of T cells with nucleic acid encoding TCRs identified in step (a), c) exposing the plurality of transfected cells to the target antigen, and d) selecting cells having TCR activity in the presence of the target antigen. Transfection of the T cells is such that each T cell comprises a nucleic acid encoding a single TCR from the library of particles, the nucleic acid is integrated into the genome of the T cell at a single identical pre-defined locus, and the TCR is in a single chain format and comprises an alpha chain variable domain, a beta cain variable domain and a constant domain. The T cells do not express endogenous TCR. A T-cell comprising a recombinase mediated landing pad, wherein the T cell constitutively expresses TCR constant domain and does not express endogenous TCR, is also claimed.

Abstract

Specific binding molecules, including T cell receptors (TCRs), which bind the HLA-A*02 restricted peptide SLYNTVATL (SEQ ID NO: 1) derived from the HIV Gag gene product, p17 are presented. TCRs of the present invention comprise non-natural mutations within the alpha and/or beta variable domains relative to a native TCR. The specific binding molecules of the invention have improved stability and/or yield and yet unexpectedly retain the advantageous properties of the specific binding molecules from which they are derived. Such specific binding molecules are particularly useful in the development of soluble immunotherapeutic reagents for the treatment of HIV infected individuals.

Abstract

The present invention relates to binding molecules comprising a peptide-major histocompatibility complex (pMHC)-binding domain which binds to a ALWGPDPAAA (SEQ ID NO: 1) peptide (derived from human pre-pro insulin (PPI) protein) presented as a peptide-HLA-A*02 complex. Said binding molecules may comprise CDR sequences embedded within a framework sequence. The CDRs and framework sequences may correspond to a T cell receptor (TCR) variable domain and may further comprise non-natural mutations relative to a native TCR variable domain. The binding molecules of the invention have high affinity and high specificity for the ALWGPDPAAA (SEQ ID NO: 1)-HLA-A*02 complex. The binding molecules may further comprise an immune suppressor and/or a half-life extending domain. Such binding molecules are particularly useful in the development of soluble immunotherapeutic reagents for the treatment of autoimmune diseases, such as diabetes.

Abstract

The present invention relates to specific binding molecules, such as T cell receptors (TCRs), that bind the HLA-A*02 restricted peptide KVLEYVIKV (SEQ ID NO: 1) derived from the germline cancer antigen MAGEA1. The specific binding molecules may comprise non-natural mutations within the alpha and/or beta variable domains relative to a native MAGEA1 TCR. The specific binding molecules of the invention are particularly suitable for use as novel immunotherapeutic reagents for the treatment of malignant disease.

Abstract

The present invention relates to multi-domain binding molecules. The molecules comprise i) a peptide-major histocompatibility complex (pMHC) binding domain comprising a first variable region linked to a constant region (VC1) and a second variable region linked to a constant region (VC2), wherein VC1 and VC2 dimerise to form the pMHC binding domain; ii) an immune cell engaging domain comprising an antibody light chain variable domain (TCE-VL) linked to an antibody heavy chain variable domain (TCE-VH); and iii) a half-life extending domain comprising a first portion of an IgG Fc region (FC1) and a second portion of an IgG1 Fc region (FC2); wherein the multi-domain binding molecule comprises:a first polypeptide chain in which the immune cell engaging domain is linked to the N terminus of VC1; a second polypeptide chain in which VC2 is linked via its C terminus to the N terminus of FC1; and a third polypeptide chain comprising FC2; and wherein the pMHC binding domain and the immune cell engaging domain are capable of binding to a pMHC complex and a T cell respectively. The binding molecules can be used to treat diseases such as cancer, autoimmune diseases and infectious diseases.

Abstract

The present invention relates to specific binding molecules that bind the HLA-A*02 restricted peptide GLSPTVWLSV (SEQ ID NO: 1) derived from HBV envelope protein. The specific binding molecules may comprise alpha and/or beta TCR variable domains and may comprise non-natural mutations within the alpha and/or beta variable domains relative to a native TCR. The specific binding molecules of the invention are particularly suitable for use as novel immunotherapeutic reagents for the treatment of infectious or malignant disease.
