Abstract

The present disclosure concerns a Reg3α polypeptide (also known as Hepatocarcinoma-Intestine-Pancreas/Pancreatitis Associated Protein (HIP/PAP)) for protecting oxygen sensitive gram-positive bacteria, compositions including the polypeptide, and their use. The disclosure shows that increasing concentration of the hReg3α lectin into the gastrointestinal tract (GIT) lumen of hReg3α-transgenic mice induced significant changes in composition of gut microbiota, and dramatically improved host resistance to intestinal inflammation. The Reg3α polypeptide may be used for preventing or treating microbiota-related diseases and/or disorders, including inflammatory bowel disease (IBD), colitis, gastrointestinal infections, irritable bowel syndrome and other gastrointestinal functional diseases, gastrointestinal tract cancer, metabolic syndrome and obesity, diabetes, liver diseases, allergic diseases, neurodegenerative diseases and psychological disorders, and the prevention and treatment methods may also include transferring to said patient fecal microbiota from a human or animal expressing the Reg3α polypeptide.

Abstract

The Reg3α polypeptide may be used for preventing or treating microbiota-related diseases and/or disorders, including inflammatory bowel disease (IBD), colitis, gastrointestinal infections, irritable bowel syndrome and other gastrointestinal functional diseases, gastrointestinal tract cancer, metabolic syndrome and obesity, diabetes, liver diseases, allergic diseases, neurodegenerative diseases and psychological disorders.

Abstract

F.prausnitziiprausnitzii and were shown as deriving from a single Microbial Anti- inflammatory Molecule (MAM). The inventors shows that L-MAM treatment prevented animals from development of High-Fat Diet- induced obesity. L-MAM treated mice significantly gained less weight throughout the gavage as compared to HFD-fed control experiment. The difference in body weight between L-MAM-treated animals on a HFD and the remaining animals is largely due to a significant reduction in the percentage of fat mass and a significant increase in the percentage of lean mass. Oral glucose tolerance testing (OGTT) revealed that chronic administration of L-MAM increased glucose tolerance. Finally, all adipose tissues weights were lower in L-MAM- treated mice. Accordingly, the present invention relates to the use of MAM polypeptides for the treatment of obesity and obesity-related disorders.

The present invention relates to allergy field. Several independent groups have recently investigated the implication of PCSK9 on inflammation and sepsis but none of them have determine its impact on allergies and/or asthma which is a global health burden. Inventors have obtained preliminary data on wild-type (PCSK9+/+) or PCSK9-deficient mice (PCSK9-/-) and shown that, under basal condition and in the absence of a particular stimulus, PCSK9 deficiency significantly increases the percentage of regulatory T cells in the spleen, the mesenteric lymph nodes and Peyer's patches. Moreover, inventors have shown the effect of allergic challenge on primary human bronchial epithelial cells on PCSK9 expression and secretion. Very interestingly, their first results obtained by Q-PCR showed that HDM and LPS increase PCSK9 mRNA levels. Accordingly, the present invention relates to inhibitors of PCSK9 for use in the treatment of asthma and/or allergic disease, such as food allergy.

Abstract

Immunoglobulin A nephropathy (IgAN) is the most common primary glomerulonephritis worldwide. IgA is mainly produced by the gut associated lymphoid tissue (GALT). Both experimental and clinical data suggest a role of the gut microbiota in this disease. The inventors aimed to determine if an intervention targeting the gut microbiota could impact disease development in a humanized mouse model of IgAN, the α1KI-CD89TG mice. Four- week old mice were divided into two groups to receive either antibiotics or vehicle-control by oral gavage twice a week for eight weeks. Antibiotic treatment efficiently depleted the faecal microbiota, impaired GALT architecture and impacted mouse IgA production. However, while hlgA1 and mIgG serum levels were unchanged, the antibiotic treatment markedly prevented hlgA1 mesangial deposition, glomerular inflammation and the development of proteinuria. This was associated with a significant decrease in circulating hlgA1-mIgG complexes. These data support that use of antibiotics would be suitable for the treatment of immunoglobulin A nephropathy.

The present invention relates to a method of treating allergic asthma in a subject in need thereof comprising a step of administering to said subject a therapeutically effective amount of a polypeptide comprising or consisting of a biologically active fragment of the house-dust mite Dermatophagoides pteronyssinus Der p 2.1. Inventors have used a model allergy in a severe asthma allergic (mouse Balbc). They have surprisingly found that when they injected twice the polypeptide derp2.1 in the mouse model after a third asthma attack, the mouse presents a respiratory improvement, reduction of neutrophils and eosinophils in the broncho-alveolar lavage (BAL), an increase of regulators lymphocytes T and reduction of natural killer cells in the BAL. Thus, the polypeptide derp2.1 is a new tool to treat the allergic asthma by a restoration of lung function and reduction of inflammatory environment.

Abstract

The present invention concerns a process for the preparation of stabilized protein aggregate particles, wherein said process comprises the following steps: a) providing an aqueous solution wherein are dispersed proteins and at least one reducing sugar, b) adjusting the pH of the aqueous solution obtained in step a), to a pH above 7, c) drying the aqueous solution obtained in step b), to form powder particles comprising said proteins and said at least one reducing sugar, d) dry heating said powder particles obtained in step c), to form said stabilized protein aggregate particles.

Abstract

The present disclosure concerns a process for obtaining a variant yeast strain capable of producing less ethanol in an alcoholic fermentation process than its corresponding ancestral strain. The variant yeast strain is obtained by culturing the ancestral strain in the presence of increasing concentrations of a salt capable of causing an hyperosmotic stress to the ancestral yeast strain. The present disclosure also concerns variant yeast strain obtained from this process (for example the variant yeast strain deposited at Institut Pasteur, on January 9, 2014, under accession number CNCM I-4832, the variant yeast strain deposited at Institut Pasteur, on October 18, 2012 under accession number CNCM I-4684, the variant yeast strain deposited at Institut Pasteur, on October 18, 2012 under accession number CNCM I-4685 and/or the variant yeast strain deposited at Institut Pasteur on January 28, 2015 under accession number CNCM I-4952) as well as processes using the variant yeast strain (wine fermentation for example).

Abstract

The present invention relates to a radioluminescent compound for radiotherapy and deep photodynamic therapy (Deep PDT), the radioluminescent compound comprising a molecular conjugate made up of a radioluminescent molecule and one photosensitiser, the radioluminescent molecule being suitable for absorbing an X-ray with energy higher than an absorption threshold and for emitting luminescent radiation in the visible domain, and the photosensitiser being suitable for absorbing said luminescent radiation and producing singlet oxygen. According to the invention, the radioluminescent compound is made up of a molecule of lanthanide chlorine (LnCl3); said photosensitiser is selected among the following photosensitisers: Al(III)phthalocyanine, m-THPC, chlorin e6 (Ce6), hypericin, hypocrellin, Nile blue, oxazine 170, oxazine 1, protoporphyrin IX, 7-methoxycoumarin-4-acetic acid, bacteriochlorophyll, and auramine; and said photosensitiser is selected such as to maximise the energy transfer between an X-ray absorbed by the radioluminescent lanthanide 2 and the photosensitiser in order to produce singlet oxygen.

Abstract

The present invention relates to a process for the production of a powdered dry emulsion containing at least one lipophilic active ingredient, intended for improving the bioavailability of said lipophilic active ingredient, which comprises the following steps: (a) a step of providing an aqueous phase containing at least one emulsifying compound which is protein in nature, chosen from whey proteins, and a lipid phase containing said lipophilic active ingredient(s), (b) a step of forming an oil-in-water emulsion, (c) a step of incorporating at least one saccharide compound into said oil-in-water emulsion resulting from step (b), and (d) a step of eliminating said aqueous phase in order to obtain said powdered dry emulsion.

Abstract

The present invention relates to the field of probiotics. More particularly, the present invention relates to a probiotic Lactobacillus plantarum bacterial strain which has a positive effect on the immune system, both in terms of innate immunity and in terms of adaptive immunity. This effect is manifested in particular in the case of influenza. Even more surprisingly, this bacterial strain exhibits resistance to bile salts, which are a stress inherent in the human digestive tract and in particular in the intestines. The present invention also relates to uses of said bacterial strain. Moreover, the present invention relates to a composition containing said strain.

Abstract

The invention relates to a combination product including, as active substances, at least one inhibitor of the HMG-CoA reductase enzyme and citrulline or a compound that is the bioequivalent thereof, for treating excess weight or obesity and/or the accumulation of body fat.
