Abstract

Hepatitis D virus (HDV) infection is the most severe form of chronic viral hepatitis due to rapid progression towards liver-related death and hepatocellular carcinoma. Now the inventors have demonstrated that CYP3A4 inhibitors can be useful for the treatment of HDV infections. In particular, the inventors showed that ritonavir a very potent CYP3A4 inhibitor had a significant impact on the secretion of HBs. The effects were confirmed with Saquinavir and Ketoconazole on the secretion of HBs. Finally, the inventors showed that ritonavir has a limited effect on the replication of HBV and HDV in coinfected cells, but dramatically reduced the number of infectious particles in culture supernatants. Accordingly, the present invention relates to the use of CYP3A4 inhibitors for the treatment of hepatitis D virus (HDV) infections.

Abstract

The present invention relates to a method of determining metabolic parameters of one or several muscular motor units by coupling muscular response to ventilatory response of a subject, comprising, from a step of performing efforts (i) using a test device by the subject moving on the test device; (ii)-a step of instantaneously measuring the movement speed of one or several muscular motor units of the subject v(t), and the oxygen flow rate φO2t); (iii)-a step of plotting the COT curve of the oxygen flow rate of the subject against the movement speed of one or several muscular motor units of the subject v(t), (iv)-a step of determining the following metabolic parameters from the COT curve: Basal power B which is the vertical asymptote at the origin, Flow resistance RM which is the oblique asymptote at high intensities.

Abstract

The present invention relates to nuclease protein fusions for enhancing genome editing by homology-directed transgene integration (HDI). The inventors found that the rate of HDI mediated by the CRISPR/Cas9 system may be substantially improved by providing the Cas9 nuclease in the form of a fusion protein with at least the N-terminal domain of the CtIP protein. CtIP proteins are involved in the early steps of homologous recombination. In addition, the inventors identified the subdomains of the N-terminal domain of the CtIP protein that are important for improving the HDI rate. Thus, the invention relates to fusion proteins comprising a Cas9 protein, a tetramerization domain of a CtIP protein and a dimerization domain of a CtIP protein. Particularly, the inventors have tested these fusion proteins HEK293 cells, RG37DR cells and Sprague-Dawley rats.

Abstract

The invention concerns the use of Lepista flaccida intended for treating a disease caused by a nonsense mutation of a gene resulting in the premature introduction of a UGA and/or UAA stop codon. It also concerns a Lepista flaccida extract comprising at least 2,6-diaminopurine (DAP) obtained by maceration or infusion of Lepista flaccida in a solvent. It also concerns this Lepista flaccida extract intended to be used for treating a disease caused by a nonsense mutation of a gene resulting in the premature introduction of a UGA and/or UAA stop codon. It additionally concerns a therapeutic treatment method comprising a step of administering Lepista flaccida or a Lepista flaccida extract or a pharmaceutical composition comprising this Lepista flaccida extract. The invention is applicable in the pharmaceutical field, in particular for treating diseases caused by a nonsense mutation of a gene resulting in the premature introduction of a UGA and/or UAA stop codon.

Abstract

The invention relates to a method of generating control commands for coordinating displacement members of a moving platform, so as to make said moving platform progress between an initial speed and a final speed according to at least two locomotive situations. The method of the invention is characterized in that each locomotive situation is associated with an elementary module (Mk) corresponding thereto for generating commands for controlling coordination between a predetermined module start speed and a predetermined module end speed, and in that domains of application of these elementary modules are represented in the form of a predetermined chart for stringing together adjoining modules covering at least partially, by juxtaposition of their start and end speeds, the spectrum of possible initial and final speeds of displacement of the platform, so as to allow the determination (330) of a series of elementary modules to be strung together to pass from the initial speed (Vi) to the final speed (Vf) and the concatenation (340) of the modules of this series so as to derive therefrom a series of corresponding control commands (350) for the displacement members for the moving platform.

Abstract

The present invention relates to a fish protein hydrolysate containing molecules capable of exerting a satietogenic activity and of regulating food intake in humans or animals. More specifically, the protein hydrolysate according to the invention enables stimulation of the secretion of endogenous cholescystokinins (CCKs) and of endogenous glucagon-like peptide 1 (GLP1) molecules by intestinal cells and the supply of exogenous CCKs. The fish protein hydrolysate according to the invention is obtained by enzymatic hydrolysis of at least one protein source selected from the group composed of the pelagic fish species Micromesistius poutassou, Clupea harengus, Scomber scombrus, Sardina pilchardus, Trisopterus esmarki and Trachurus spp., the demersal fish species Gadus morhua, Pollachius virens, Melanogrammus aeglefinus and Coryphaenoides rupestris, and the species of fish belonging to the order Siluriformes, said enzymatic hydrolysis being carried out by means of a mixture of enzymes comprising endopeptidases derived from Bacillus amyloliquefaciens and from Bacillus licheniformis, or derived from Bacillus amyloliquefaciens, from Bacillus licheniformis and from Aspergillus oryzae.

Abstract

The present invention relates to an anti-gene oligonucleotide which specifically hybridizes with the gene coding for the human tyrosinase related protein-1 by Hoogsteen pairing between complementary bases, said oligonucleotide forming a triple helix structure with the human tyrosinase related protein-1 gene. The invention also relates to the use of said oligonucleotide as skin depigmenting or whitening agent in a cosmetic composition or a dermatological composition.

Abstract

The invention relates to compositions and methods for interfering with the DNA repair of double strand breaks (DSBs). The invention discloses novel double-stranded nucleic acid molecules that act as baits and hijack the holocomplex of enzymes responsible of DNA DSB sensing, signaling and/or repair pathways, in particular the non homologous end joining (NHEJ) pathway of DSB repair. The invention discloses the use of these molecules as adjuvant compositions to be used in association with a DNA breaking treatment, particularly radiotherapy or chemotherapy, in combination with a pharmaceutically acceptable carrier, in an efficient amount to be introduced in the tumor cell nuclei in order to neutralize transiently their DNA repair capacity and trigger their death.

Abstract

The invention relates to double-stranded nucleic acid fragments comprising a chemically modified backdone and at least 4-1000 bp, preferably 8-500 bp, and most preferably 16-200 bp. The disclosed molecules (DRIL molecules) may interfere with DNA damage signalling and repair pathways, in particular the non homologous NHEJ pathway of double-stranded break repair. The invention discloses the application of the DRIL molecules as adjuvant compositions to be used in association with a DNA breaking treatment, particularly radiotherapy or chemotherapy, in combination with a pharmaceutically acceptable carrier, in an efficient amount to be introduced in the tumoral cell nuclei in order to trigger DNA repair induced lethality (DRIL in short) of tumoral cells/tissues.
