A technique is provided to visualize microtubules in live cells that does not require genetic manipulation or microinjection. Moreover, this method also avoids perturbation of the endogenous microtubule network that occurs with taxol treatment. This technique exploits tyrosination and detyrosination of tubulin, a posttranslational modification cycle specific to the C-terminus of a- tubulin. Specifically, cells are grown in medium supplemented with a tyrosine derivative possessing a reactive functional group. The cellular enzyme tubulin tyrosine ligase attaches the unnatural amino acid to a single site on tubulin. Addition of fresh medium containing a suitably derivatized fluorophore then yields fluorescent tubulin, which incorporate into cellular microtubules. Importantly, the tubulin labeling approach demonstrated here does not detrimentally affect microtubule network or cell morphology. Thus we present a simple, robust labeling technique that allows microscopic analysis of microtubules in live cells.

The present invention provides peptides (including analogs and derivatives thereof) corresponding to residues 96-110 and 35-47 of ras-p21, which peptides have attached thereto a membrane-penetrating leader sequence. The subject peptides, analogs and derivatives thereof are useful in treatment of cancers and have been shown to induce phenotypic reversion of pancreatic cancer cells to non-cancerous cells. Pharmaceutical compositions comprising one or more subject peptides are also provided by the present invention. The present invention further provides replication incompetent Adenovirus (AdV) vectors comprising a promoter sequence and a nucleotide sequence encoding a subject peptide. Methods of treating cancer by administering one or more subject peptides, pharmaceutical compositions, and/or AdV vectors are also provided.

There is a constant battle to break even between continuing improvements in DRAM capacities and the growing memory demands of large-memory high-performance applications. Performance of such applications degrades quickly once the system hits the physical memory limit and starts swapping to the local disk. We present the design, implementation and evaluation of Anemone—an Adaptive Network Memory Engine—that virtualizes the collective unused memory of multiple machines across a gigabit Ethernet LAN, without requiring any modifications to the either the large memory applications or the Linux kernel. We have implemented a working prototype of Anemone and evaluated it using real-world unmodified applications such as ray-tracing and large in-memory sorting. Our results with the Anemone prototype show that unmodified single-process applications execute 2 to 3 times faster and multiple concurrent processes execute 6 to 7.7 times faster, when compared to disk based paging. The Anemone prototype reduces page-fault latencies by a factor of 19.6—from an average of 9.8 ms with disk based paging to 500 μs with Anemone. Most importantly, Anemone provides a virtualized low-latency access to potentially “unlimited” network memory resources.
