Disclosed is DNA valency sorting chromatography, a purification method for separating solutes based on the number of barcoded DNA molecules present on their surface, which can operate using conventional low-pressure chromatography equipment and instrumentation. Microbial consortia exert great influence over the physiology of humans, animals, plants, and ecosystems. The approach disclosed herein provides an effective method to dynamically control population compositions in microbial consortia, which we demonstrate in the context of co-culture fermentations for chemical production. Co-culture fermentations can improve chemical production from complex biosynthetic pathways over monocultures by distributing enzymes across multiple strains, thereby reducing metabolic burden, overcoming endogenous regulatory mechanisms, or exploiting natural traits of different microbial species. However, stabilizing and optimizing microbial sub-populations for maximal chemical production remains a major obstacle in the field. An optogenetic circuit, called OptoTA, is disclosed for regulating a toxin-antitoxin system, which enables tunability of, e.g., Escherichia coli growth using only blue light. With the disclosed system, one can control population ratios of co-cultures of, e.g., E. coli and Saccharomyces cerevisiae containing different metabolic modules of biosynthetic pathways. The electron donor, in some embodiments, comprises ammonium, an ammonium containing compound, and/or molecular hydrogen. A two-step process is provided for forming large photonic single crystals of about 0.1 millimeter and greater via DNA coated colloidal particles. The two-step process generally include decoupling the nucleation and growth steps. In particular, DNA colloidal particles are partitioned in nanoliter droplets formed in a water in oil emulsion using microfluidics. Once a crystal nucleates within a droplet, depletion of particles occurs as the crystal grows inhibit formation of more crystals within the droplet. A small number of droplets containing these seed crystals are then mixed with droplets containing weak DNA coated colloidal particles. The emulsion is then broken and heated at a temperature effective to cause dissociation of the weak particles while the seeds remain stable. The system is further cooled at a temperature effective that the particles stably adhere to the seed crystals resulting in growth while inhibiting nucleation of new crystals.

Disclosed are criteria for a lattice matched, multilayer, organic crystal heterostructure. Current organic devices (e.g., photovoltaics, light emitting diodes, or transistors) rely on amorphous material despite superior charge transport properties of crystalline organic semiconductors. Achieving a fully crystalline device architecture requires growth of a molecular crystal atop a different one, or heteroepitaxy, and is particularly relevant in organic semiconductor devices that demand multiple layers of different molecules. This challenge is complicated when attempting to stack highly ordered layers needed for crystalline devices because strategies are needed to ensure that each layer grows crystalline. It is shown herein that lattice matching alone is not sufficient for successful organic heteroepitaxy deposited via physical vapor deposition. The process disclosed herein includes an additional criterion in which the lattice matched plane of the adlayer must also be the crystal face with the lowest surface energy. Application of this process leads to a full crystalline multilayer system in which there is perfect registry between the template layer and adlayer. Not only does this allow for the study of highly ordered interfaces, but it also opens the door to entirely crystalline device architectures, likely improving the efficiency over their amorphous counterparts.
