Building strong underwater bioadhesion is important for several applications but proved to be an extremely challenging task. In a wet/underwater environment, the water boundary layer hinders interfacial adhesion, which is further undermined by water-induced swelling in bioadhesives. In contrast to current hydrophilic bioadhesives, we here propose an underwater and in-situ applicable hydrophobic adhesive (UIHA). We report polydimethylsiloxane (PDMS) tangled with macromolecular silicone fluid as a dissipation phase to provide strong interface adhesion strength. The silicone fluid repels the surface boundary water layer, overcomes capillary break up in water, and rapidly gelates with PDMS, while a small amount of silane (<0.2%) can bridge the hydrophobic adhesive and tissue substrates to an exceptional underwater adhesive strength. UIHA presents in-situ and instant adhesive performances when tested on artery, lung, bone, and skin tissues.

A biosensor for real-time monitoring of a wound can provide valuable data regarding the presence of bacteria. A nanofibrous biosensor is provided for monitoring wound status at the point of care. The colorimetric biosensor changes color in response to low levels of bacteria and fungi, making it visible to unaided and untrained eyes. A colorimetric probe is a hemicyanine dye that changes color from yellow to green in the presence of lipase. Dye is incorporated into a shell composition of core-shell nanofibers made of polyurethane and polyvinylpyrrolidone. As a means of increasing the biosensor's sensitivity, the alignment of nanofibers is controlled and a surfactant added to the shell (e.g., Tween 80). The procedure is most useful for sparse matrix equations, as it allows for the inverse of a matrix to be applied with (Nnz log(N)) complexity, where N is the number of unknowns, and Nnz is the total number of nonzero elements in the system matrix. This efficiency is independent of the matrix structure, and hence the quantum procedure can outperform classical methods for many common system types. We show this using the example of sparse approximate inverse (SPAI) preconditioning, which involves the application of matrix inverses for matrices with Nnz=(N).

Multiple sclerosis (MS) is characterized by immune mediated neurodegeneration that results in progressive, life-long neurological and cognitive impairments. Yet, the endogenous mechanisms underlying MS pathophysiology are not fully understood. Here, we provide compelling evidence that associates dysregulation of neuregulin-1 beta 1 (Nrg-1β1) with MS pathogenesis and progression. In the experimental autoimmune encephalomyelitis (EAE) model of MS, we demonstrate that Nrg-1β1 levels are abated within spinal cord lesions and peripherally in the plasma and spleen during presymptomatic, onset and progressive course of the disease. We demonstrate that plasma levels of Nrg-1β1 are also significantly reduced in individuals with early MS and is positively associated with progression to relapsing-remitting MS. The functional impact of Nrg-1β1 downregulation preceded disease onset and progression, and its systemic restoration was sufficient to delay EAE symptoms and alleviate disease burden. Intriguingly, Nrg-1β1 therapy exhibited a desirable and extended therapeutic time window of efficacy when administered prophylactically, symptomatically, acutely or chronically. Using in vivo and in vitro assessments, we identified that Nrg-1β1 treatment mediates its beneficial effects in EAE by providing a more balanced immune response. Mechanistically, Nrg-1β1 moderated monocyte infiltration at the blood-central nervous system interface by attenuating chondroitin sulfate proteoglycans and matrix metalloproteinase-9. Moreover, Nrg-1β1 fostered a regulatory and reparative phenotype in macrophages, T helper type 1 (Th1) cells and microglia in the spinal cord lesions of EAE mice. Taken together, our new findings in MS and EAE have uncovered a novel regulatory role for Nrg-1β1 early in the disease course and suggest its potential as a specific therapeutic target to ameliorate disease progression and severity.

Screening for novel antibacterial compounds in small molecule libraries has a low success rate. Here, we applied a machine learning (ML)-based virtual screening for antibacterial activity and evaluated its predictive power by experimental validation. We first binarized thirty-thousand compounds according to their growth inhibitory activity (hit rate 0.67%) against the antibiotic-resistant bacterium Burkholderia cenocepacia and described their molecular features with a directed-message passing neural network (D-MPNN). Then, we used the data to train an ML model that achieved a receiver operating characteristic (ROC) score of 0.823 on the test set. Finally, we predicted antibacterial activity in virtual libraries corresponding to 1,614 compounds from the Food and Drug Administration (FDA)-approved list and 224,205 natural products. A hit rate of 26% and 12%, respectively, was obtained when we tested the top ranked predicted compounds for growth inhibitory activity against B. cenocepacia, which represents at least a 12-fold increase from the previous hit rate. In addition, more than 51 % of the predicted antibacterial natural compounds inhibited ESKAPE pathogens showing that predictions expand beyond the organism-specific dataset to a broad range of bacteria. Overall, the developed ML approach can be used for compound prioritization prior to screening, increasing the typical hit rate of high throughput screens. Also described are anti-bacterial compounds isolated by the screen.

Acoustic imaging is performed, and prior information extracted from the sound-speed image is incorporated into the electromagnetic inversion algorithm, whereby low-resolution sound-speed reconstructions are used to improve tumour detection via electromagnetic imaging.
An electromagnetic imaging method uses a reduced set of whole domain basis functions that is chosen in an informed manner from prior information about the imaging problem, using prior information to tailor the choice of 'alternative' basis functions, e.g., first seeking layered structure in a grain storage bin, then providing support for hot-spots. In one embodiment, the method first uses a pulse basis and then projects the basis onto the 'alternative' basis. This allows for more than one 'alternative' basis at each iteration. Similarities and differences between the target reconstruction in different alternative bases can be used to guide the reconstruction procedure. Described herein is a strategy for converting 2D transition-metal titanium carbide MXene (M3X2Tx) sheets to multi-dimensional GerMXene superlattice heterostructure. The GerMXene possesses a unique microstructure with enhanced surface properties compared to its parent materials, MXene and Xene (GeH). Also, GerMXene offers excellent biocompatibility and bioactivity with tailored electrical and optical properties for application in multiple fields.
Building strong underwater bioadhesion is important for several applications but proved to be an extremely challenging task. In a wet/underwater environment, the water boundary layer hinders interfacial adhesion, which is further undermined by water-induced swelling in bioadhesives. In contrast to current hydrophilic bioadhesives, we here propose an underwater and in-situ applicable hydrophobic adhesive (UIHA). We report polydimethylsiloxane (PDMS) tangled with macromolecular silicone fluid as a dissipation phase to provide strong interface adhesion strength. The silicone fluid repels the surface boundary water layer, overcomes capillary break up in water, and rapidly gelates with PDMS, while a small amount of silane (<0.2%) can bridge the hydrophobic adhesive and tissue substrates to an exceptional underwater adhesive strength. UIHA presents in-situ and instant adhesive performances when tested on artery, lung, bone, and skin tissues.
Described herein is a biocompatible lipid nanoparticle (LNP) composition suitable for delivering RNA payloads into cells and tissues of a subject. A rapid, low-cost, portable and reliable method for on-site detection of deoxynivalenol (DON), a representative mycotoxin predominantly occurring in grains, would be helpful to control mycotoxin contamination. Herein, a paper-based microfluidic chip capable of measuring deoxynivalenol (DON-Chip) in foods, feeds and feed ingredients was developed. As discussed herein, the DON-Chip incorporated a colorimetric competitive immunoassay into a paper microfluidic device and used gold nanoparticles as a signal indicator. Furthermore, a novel ratiometric analysis method was used to improve signal resolvability at low concentrations of DON. Detection of DON in aqueous extracts from solid foods, feeds or feed ingredients was successfully validated with a detection range from 0.01-20 ppm (using dilution factors from 10-104). Compared with conventional methods, the novel DON-Chip greatly reduces the cost and time of mycotoxin detection in the food and feed industry.

Homodimeric Tobramycin Adjuvant Repurposes Novobiocin as an Effective Antibacterial Agent Against Gram-Negative Bacteria
Low permeability across the outer membrane is a major reason why most antibiotics are ineffective against Gram-negative bacteria. Agents that permeabilize the outer membrane are typically toxic at their effective concentrations. Here, we report the development of a broad-spectrum homodimeric tobramycin adjuvant that is non-toxic and more potent than the gold standard permeabilizing agent, polymyxin B nonapeptide. In pilot studies, the adjuvant confers potent bactericidal activity on novobiocin against Gram-negative bacteria, including carbapenem-resistant and colistin-resistant strains bearing plasmid-borne mcr-1 genes. Resistance development to the combination was significantly reduced, relative to novobiocin alone, and there was no induction of cross-resistance to other antibiotics, including the gyrase-acting fluoroquinolones. Tobramycin homodimer may allow the use of lower doses of novobiocin, overcoming its twin-problem of efficacy and toxicity.

Flea beetles (species of the genus Phyllotreta) are serious pests of cruciferous crops, causing millions of dollars losses each year, despite the use of chemical pesticides. Our current pesticides are broad-spectrum in their activity, killing not just the pests, but also many non-target species, including beneficial insects such as pollinators and predators. Here, an alternative set of pesticides, based on double-stranded RNA (dsRNA), is shown to be effective at killing flea beetles when the dsRNAs are applied to a leaf surface and fed to the insects. Insects that fed on leaves sprayed with the dsRNAs died within 8 days of first exposure, but also showed reduced feeding activity within several days, thereby reducing feeding damage to the plants. Importantly, the dsRNAs were designed to be specific for the flea beetles, and when delivered to non-target beetles, no adverse effects were observed, illustrating the specificity of this new type of pesticide.

Using Panx1 deletion variants, we identified the STIM interaction domain within the intracellular N-terminus of Panx1 (amino acids 1-18), which is necessary for promoting Panx1 activation. Further, in hippocampal cultured neurons we show that ionotropic function of NMDARs causes Panx1 activation and knockdown of STIM inhibits Panx1 activation. Panx1-STIM region controls NMDAR-initiated Panx1 activation confirming the model of NMDAR-STIM-Panx1 signalling. N-term (amino acids 19-35) is responsible for plasma membrane trafficking of Panx1 channels. The STIM interaction region of Panx1 is at amino acids 13-18. This region is required to stimulate the pathological activation of Panx1, a response linked to detrimental actions of amyloid beta. This technology provides a target to treat Alzheimer's disease.

USE OF NRG-1BETA1 FOR DETECTION AND/OR TREATMENT OF MULTIPLE SCLEROSIS
Multiple sclerosis (MS) is characterized by immune mediated neurodegeneration that results in progressive, life-long neurological and cognitive impairments. Yet, the endogenous mechanisms underlying MS pathophysiology are not fully understood. Here, we provide compelling evidence that associates dysregulation of neuregulin-1 beta 1 (Nrg-1β1) with MS pathogenesis and progression. In the experimental autoimmune encephalomyelitis (EAE) model of MS, we demonstrate that Nrg-1β1 levels are abated within spinal cord lesions and peripherally in the plasma and spleen during pre-symptomatic, onset and progressive course of the disease. We demonstrate that plasma levels of Nrg-1β1 are also significantly reduced in individuals with early MS and is positively associated with progression to relapsing-remitting MS. The functional impact of Nrg-1β1 downregulation preceded disease onset and progression, and its systemic restoration was sufficient to delay EAE symptoms and alleviate disease burden. Intriguingly, Nrg-1β1 therapy exhibited a desirable and extended therapeutic time window of efficacy when administered prophylactically, symptomatically, acutely or chronically. Using in vivo and in vitro assessments, we identified that Nrg-1β1 treatment mediates its beneficial effects in EAE by providing a more balanced immune response. Mechanistically, Nrg-1β1 moderated monocyte infiltration at the blood-central nervous system interface by attenuating chondroitin sulfate proteoglycans and matrix metalloproteinase-9. Moreover, Nrg-1β1 fostered a regulatory and reparative phenotype in macrophages, T helper type 1 (Th1) cells and microglia in the spinal cord lesions of EAE mice. Taken together, our new findings in MS and EAE have uncovered a novel regulatory role for Nrg-1β1 early in the disease course and suggest its potential as a specific therapeutic target to ameliorate disease progression and severity.

We developed a method using monoclonal antibodies (mAbs) targeting different protein domains of FRAS-related extracellular matrix protein 1 (FREM1) to detect and quantify soluble toll-like interleukin-1 receptor regulator (sTILRR) in human plasma samples. We used affinity purification and Western blot analysis to validate the results. Using these methods we quantified sTILRR in the plasma samples. Our study showed that TILRR exists as a soluble protein in human blood plasma that can be quantified.

Described herein is a "paperclip" structured dsRNA (pcRNA) that has two closed ends (Figure 1), and was developed and tested for its ability to enter insect cells and induce RNAi. While conventional dsRNAs, with their two collinear RNA strands, enter insect cells by clathrin-mediated endocytosis, the pcRNAs can enter cells by a clathrin-independent mechanism. The new structured dsRNA can be used to deliver dsRNA to insects that either develop resistance through alterations to their conventional uptake mechanisms and to insects that are naturally refractory to dsRNAs, including lepidopteran pests.
An ultrasonic sensing technique and a signal interpretation method based on a spectral element multiphase poromechanical approach overcomes critical gaps in permafrost characterization. Ultrasonic sensing produces high-quality response signals that are sensitive to the permafrost properties. A transfer function denoting a ratio of induced displacement and applied force in the frequency domain, is independent of the distribution of the stress force applied by the transducer to the permafrost sample, and allows interpretation of the measured electrical signal using a theoretical transfer function relation to efficiently determine the most probable permafrost properties from response signals using an inverse spectral element multiphase poromechanical approach. This ultrasonic sensing technique enables rapid characterization of permafrost samples in terms of both physical and mechanical properties. The Quantitative Ultrasound (QUS) system can be used in a laboratory setup or brought on site for in-situ investigation of permafrost or frozen soil samples.
