Systems and methods for producing bioproducts are shown. The systems and methods herein can be configured and used in an integrated biorefinery. The integrated biorefinery may comprise a sugar production facility such as a sugar mill, a production facility for one or more bioproduct(s) such as butanol, and optionally an ethanol production facility employing the system and method.
Methods are provided for preparing a hydrolysate containing soluble sugar molecules from biomass that contains cellulose and hemicellulose. Hemicellulose sugars are extracted in the process, and the resulting hydrolysate may be used to support microbial fermentation to produce products of interest.
Methods are provided for conditioning a cellulosic biomass hydrolysate to remove or inactivate inhibitors of microbial growth and/or metabolite production. The methods include precipitation or inactivation of inhibitors with calcium hydroxide and potassium hydroxide.

Methods are provided for selecting microbial strains with improved properties for fermentation and/or bioproduct production. A salt selection is employed to identify mutants with improved bioproduct tolerance, titer, or osmotic tolerance relative to a microbial strain from which they were derived.

INTEGRATED SYSTEM AND PROCESS FOR BIOPRODUCT PRODUCTION
Processes and systems for production of bioproducts such as biofuels are provided. The bioproduct production processes and systems utilize pretreatment of a carbohydrate-containing feedstock to produce soluble sugar molecules and continuous conversion of the pretreated feedstock to a bioproduct by an immobilized fermenting microorganism.
The invention provides systems and methods of vapor compression distillation thermodynamic control. Variable control elements, such as a variable speed compressor or a heater are controlled to provide desired thermodynamic conditions for a portion of a vapor compression distillation unit. Desired thermodynamic conditions are determined based on optimization goals. Sensors provide feedback to a controller that controls that variable control elements.
The invention provides systems and methods for multistage vapor compression distillation (VCD). A plurality of VCD units may be combined to form a VCD device where heat from a prior VCD unit may be transferred to a subsequent VCD unit. Each VCD unit may have an evaporator stage and a condenser stage where heat from the condenser stage of a prior VCD unit may be transferred to the evaporator stage of the subsequent VCD unit. The amount of heat transfer between units may be controlled. A multistage VCD device may be used to separate an acetone-butanol-ethanol fermentation broth.

This invention provides compositions and methods for monitoring and regulating the production of a target product of a biochemical pathway in an organism, such as butanol. A gene encoding a light-emitting reporter molecule, such as luciferase, is operatively linked with a transcription regulatory nucleotide sequence that regulates transcription of an enzyme in the pathway that signals the rate of production of the target product, such as butanol dehyrogenase. When a microorganism is transfected with such a reporter construct and cultured, the reporter is expressed contemporaneously with the enzyme. The amount of light produced by the reporter indicates amount of enzyme being produced which, in turn, signals the amount of target product being produced. When the reporter is measured in real time, it provides information that can be used to regulate culture conditions and to optimize production of the target product.
