The present invention provides an mreC mutant and use thereof in L-valine fermentative production. The mreC mutant is a protein obtained by mutating proline at position 150 in the wild-type mreC amino acid sequence into leucine. The introduction of an mreC point mutation into the microbial genome, i.e., a C-to-T mutation at position 449 of the coding region in the wild-type mreC gene nucleotide sequence, can significantly improve the production capacity and biomass of an engineering strain of a high valine production. The knock-out and introduction of related enzyme genes of the L-valine anaerobic fermentation pathway further provide proper carbon metabolic flows and reducing capacity balance control in the L-valine production process. By the coordinative regulation of multiple genes, a metabolic pathway suitable for L-valine fermentative production is formed, thus achieving a high-level synthesis of L-valine in microorganisms.

Provided are an engineering bacteria and a method for producing beta-alanine. With respect to the original bacteria, the engineering bacteria is capable of overexpressing the alpha-carboxylase of L-aspartic acid of Bacillus subtilis as shown in sequence 6, or overexpressing a polypeptide having at least 60%, preferably 80%, more preferably 90%, further preferably 95% and most preferably 98% or even 99% homology with the enzyme and having the enzyme activity.

The method is to obtain the recombinant bacterium by mutating gene lon and gene clpA in a starting bacterium; during mutation of the gene lon in the starting bacterium, C at position 1310 from the 5' end in the nucleotide sequence of gene lon in the starting bacterium is mutated to A; and during mutation of the gene clpA in the starting bacterium, T at position 1895 from the 5' end in the nucleotide sequence of gene clpA in the starting bacterium is mutated to G. In the present invention, the recombinant bacterium XZ-A47 is obtained by mutating the gene lon and the gene clpA in the engineered Escherichia coli bacterium XZ-A26.
Disclosed are a bacterial strain capable of producing L-alanine and being tolerant to tap water, and a construction method therefor. Provided is a method for constructing a recombinant bacterium, comprising mutating a clpA gene in a starting bacterium to obtain the recombinant bacterium; during mutation of the clpA gene in the starting bacterium, T at position 1895 from the 5' end of a nucleotide sequence of the clpA gene in the starting bacterium is mutated into G. The recombinant bacterium XZ-A45 obtained by the present invention can produce large amounts of L-alanine in a fermentation culture medium formulated with tap water.
