Disclosed are methods for constructing disulfide mimetics for late-stage cyclization of peptides. The disclosed method provides late-stage cyclization via thioacetalization of peptides containing two cysteine residues to form thioacetal group. These bonds are stable under acidic, basic, and reduced conditions. The reaction can use a broad range of cyclic ketones even acetone as crosslinkers. Trifluoroacetic acid (TFA) alone can act as both the catalyst and the sole solvent, thus allowing most peptide sequences to be cyclized via the disclosed method. GENERATIVE ADVERSARIAL NETWORK-BASED LOSSLESS IMAGE COMPRESSION MODEL FOR CROSS-SECTIONAL IMAGING
A novel medical image generation approach synthesizes thin-cut computerized tomography (CT) images from thick-cut CT ones as inputs. First thick-cut CT images are obtained by maximizing the pixel-wise intensity of five or more than five continuous thin-cut ones after image registration. Second, the obtained thick-cut CT images are fed into a generator block which adopts an encoder-decoder architecture, where each thick-cut image is encoded into low-dimensional embedding space before decoding into multiple thin-cut ones. Third, a discriminator focuses on distinguishing original real thin-cut images from synthetic thin-cut images. An adversarial mechanism between the generator and discriminator causes the discriminator's output to provide an effective gradient update of the network parameters for the generator to increasingly improve the generator's ability to synthesize higher-quality thin-cut images and in turn promotes the discriminator's discriminating capability.

A portable, miniaturized microfluidic droplet-based digital polymerase chain reaction (PCR) device is provided. The device includes a droplet generation layer with a flow-focusing channel and multiple capillary channels, in which the flow-focusing channel has a flow focusing structure to generate droplets from a provided aqueous sample and extract oil, and the multiple capillary channels is configured to perform an oil extraction. An incubation layer includes heating plates, a microfluidic channel, and a container for conducting a PCR thermal cycling program. Controlled by a controller and powered by a power supplier, the PCR thermal cycling program involves maintaining predefined temperatures on each heating plate sequentially. This process ensures the heating of droplets to different temperatures at distinct times, facilitating precise and efficient PCR amplification.
Platinum (II) complexes (“Pt(II) complexes”) that can emit in the red and/or deep red regions and methods of making and using thereof are described. These Pt(II) complexes can have dinuclearity with close Pt—Pt contact and close interplanar distance, allowing for high emission quantum yield, such as >50%, short emission lifetime, such as <3 μs, and high radiative decay rate, such as in the order of 105 s−1. The Pt(II) complexes disclosed herein can be included in organic light-emitting devices, such as organic light-emitting diodes (“OLEDs”), for use in commercial applications. OLEDs containing these red-emitting Pt(II) complexes can have current efficiency and/or external quantum efficiency that are comparable or higher than commercially available red emitters and prolonged operational lifetime, such as a LT95 at 1000 nits>9000 h for a red OLED.

A composite nanofiber aerogel (CNA) is formed from aramid nanofibers (ANFs) combined with polyvinyl alcohol (PVA). These nanoscale constituents of the aerogel form 3D networks with high nodal connectivity and strongly welded connectivity joints between fibrils so that the structure has high stiffness and strength compared to other polymeric aerogels and successive breakage of crosslinks at the connectivity nodes affords energy dissipation while maintaining the overall structural integrity. Abstract
The present invention provides a scalable method for achieving shape control of diamond micro-nanoparticles, comprising air oxidizing diamond micro-nanoparticles grown by chemical vapor deposition and/or diamond micro-nanoparticles grown by high pressure and high temperature. The present invention achieves the controllable morphology transformation of diamond micro-nanoparticles via air oxidation treatment. It has been demonstrated that a series of unique shapes, including “flower” shaped, “hollow” structured, “pyramid” patterned on the surface, and “boomerang” shaped, can be achieved by altering the air oxidation parameters, i.e., temperature and duration. The scalable production of these differently shaped diamond micro-nanoparticles represents a significant scientific breakthrough together with a high commercial value. Disclosed is a computer-implemented three-dimensional image classification system (CIS) for processing and/or analyzing non-contrast computed tomography (CT) medical imaging data. The CIS is a deep neural network containing multiple Convolutional Block Attention Module (CBAM) blocks, which contain convolutional layers for feature extraction followed by CBAMs. The CBAM applies channel attention to highlight more relevant features and spatial attention to focus on more important regions. Max pooling layers operably link adjacent pairs of CBAM blocks. The output of the final CBAM block is passed to two terminal fully connected layers to generate a diagnosis. Live attenuated viruses for protection against the novel coronavirus Sars-CoV-2 are provided. The live attenuated chimeric virus strains are based on a live attenuated influenza A or B virus (LAIVA/B), used a master backbone, which includes deletion of the viral virulence element, the NS1 (non-structural protein 1) (DeLNS1), engineered to express one or more antigens of the Sars-CoV-2 (herein, CoV2Ag). The chimeric virus strain is referred to generally herein, as DelNS1-A/B-Sars-CoV-2-CoV2Ag. The DelNS1-A/B-Sars-CoV-2-CoV2Ag strain preferably shows spontaneous cold adaption with preference to grow at 30-33° C. Compositions including the chimeric virus also provided as a co-composition with a LAIVA/B. The present invention provides a diamond particle-based physical unclonable function material, and a preparation method therefor and a use thereof. The material comprises a monocrystalline silicon substrate, and diamond particles having silicon vacancy color centers and grown in situ on the monocrystalline silicon substrate. Preparation of the diamond particles on the silicon substrate relates to a random process, which is a key requirement for manufacturing a physical unclonable function label. According to the present invention, the scattering spectra, morphology features and spatial position relationship of the diamond particles are used as fingerprint information of the diamond physical unclonable function material. Due to extreme heat resistance, mechanical, chemical and light stabilities of a diamond material, the high-robustness label in the present invention can meet the requirements of many practical uses in various environments, and has practical commercial value in huge anti-counterfeit markets such as electronic components, medical packages, vehicles, and luxury goods.

Abstract
Provided is a robust nasal organoid culture system and fully characterized 2D and 3D in-vitro generated organoids, and methods of using the organoids. The nasal organoids contain a population of differentiated nasal epithelial cells having cellular heterogeneity and molecular markers similar to mature nasal epithelial tissue. Improved methods for generating two-dimensional and three-dimensional nasal organoids are also provided. The methods generally require a two-phase culture system, i.e., expansion and differentiation culture. The provided nasal organoids are suitable for recapitulating nasal tissue complexity for modeling respiratory diseases, studying barrier function and drug delivery, high-throughput drug screening, and assessing the effects of pathogens and environmental toxins on the nasal respiratory epithelium.
A rechargeable aqueous Mg battery is composed of a Mg metal anode, a solid-state aqueous polymer-based electrolyte, and an intercalation-type Prussian blue analogue cathode for ion storage. The subject invention pertains to a new method for measuring the elastic properties of microfibers by rope-coiling. Rope-coiling refers to the buckling of a slender elastic fiber caused by axial compression. A continuous flow microfluidic method enables the high-throughput measurement of the elasticity of microfibers by rope-coiling, where sample loading and unloading are not needed between consecutive measurements. In certain embodiments the coiling radius can be directly proportional to the elastic modulus of the fiber, facilitating calibration to measure fiber elasticity for high-throughput applications. Throughput can be thousands of times higher than that of a tensile tester, making possible an in situ, on-line measurement in a microfluidic production line, which couples the making of microfibers and the measurement of elasticity on the same line. The new method can also measure certain fibers with local variations in elasticity.
Genetically modified zebrafish, in which mutation combinations frequently identified in human AML are stably expressed in the stem cell population of the fish, are provided. The combination of mutations result in morphologic, cytochemical and molecular changes of its blood cells that are remarkably similar to those in human AML. The zebrafish model provides a foundation for the study of AML initiation and progression and a high throughput in vivo drug screening platform to identify personalized therapies for AML based on specific mutation combinations. A nano scale robotic system for single cell DNA sequencing of a strand of DNA positioned on a slide utilizes an atomic force microscope (AFM) having an end effector in the form of a cantilever with a tip. Abstract
Provided are engineered, replication-deficient influenza viruses that are infectious and stimulate broadly-cross protective immunity against multiple different influenza strains. Also provided are compositions and methods for providing protective immune responses against influenza. The methods deliver compositions of intact, replication-deficient influenza viruses by intradermal administration in an amount effective to elicit or stimulate a cross-protective immune response to influenza viruses in the recipient following a single administration. Because the engineered influenza viruses are non-replicating, they are safe and effective in immunocompromised subjects. Also provided are methods for delivering co-stimulatory molecules, growth factors, adjuvants and/ or cytokines together with the non-replicating influenza viruses.

Methods for asymmetric cis-dihydroxylation (“AD”) of quinones to produce cis-diols of quinones with high yield (i.e. a yield ≥30%) and high enantioselectivity (i.e. an enantiometric excess ≥30%) are disclosed. COMPOSITIONS AND METHODS FOR IMPROVED MESENCHYMAL STEM CELL THERAPY
It has been established that CD55 expression on human mesenchymal stem cells (hMSCs) is positively correlated with successful allogenic transplantation procedures. Compositions and methods for improved hMSCs, having increased expression of CD55 mRNA and/or increased expression of CD55 on the surface of the cell as compared to a control cell, are provided. Methods include isolating hMSCs from a tissue that naturally has increased CD55 expression, such as adipose tissue derived hMSC (AT-hMSC); and/or in vitro culturing hMSCs in the presence of one or more active agents that stimulates or enhances the expression of CD55 mRNA, and/or increases the surface expression of CD55 in the hMSC to provide improved hMSCs. Preferred active agents include HMG-CoA reductase inhibitors such as atorvastatin and EGFR inhibitors such as erlotinib. Abstract
An optical-resolution photoacoustic microscopy (OR-PAM) system for visualizing water content deep in biological tissue uses an all-fiber 1930-nm hybrid optical parametrically-oscillating emitter. The emitter includes a tunable laser source whose output is amplified by a first erbium-doped fiber amplifier (EDFA). The output of the first amplifier is modulated with a Mach-Zehnder amplitude modulator that receives an RF signal with a nanosecond pulse width and a multiple kilohertz repetition rate. A second EDFA further amplifies the signal and passes it to a fiber circulator that in turn delivers it to a 1950/1550 mm fiber wavelength-division-multiplexing coupler WDM. The coupler introduces the signal to a cavity that includes a spool of highly nonlinear fiber and a Thulium-doped fiber amplifier TDFA. From the TDFA the signal reaches a 50/50 fiber coupler that sends part to a second output TDFA and guides part back to the cavity through a port of the WDM.

The disclosure relates to defective interfering genes and viruses thereof. It has been discovered that defective interfering genes designed from influenza and coronavirus species can inhibit infection or replication of the parent virus. In vivo, DIGs induce rapid-onset prophylactic protection of infected animals against lethal viral doses. Thus, disclosed herein are nucleic acids containing DIGs, pharmaceutical compositions thereof and associated methods of use. For example, described herein is an isolated polynucleotide containing one or more defective interfering genes, wherein each of the one or more defective interfering genes contains a nucleotide sequence corresponding to one or more portions of an influenza or coronavirus gene or genome, wherein the nucleotide sequence includes a deletion in the gene. The DIGs can be in the form of a plasmid. Pharmaceutical compositions of the plasmid can be used to limit viral replication and prevent or treat influenza or coronavirus associated diseases.

Live attenuated viruses for protection against the novel coronavirus, designated as Sars-CoV-2 by the World Health Organization (WHO) are provided. The DelNS1-B-Sars-CoV-2-CoV2Ag strain preferably shows spontaneous cold adaption with preference to grow at 30-33° C. Methods for obtaining a population of differentiated alveolar cells, differentiated alveolar cells in the form of alveolar organoids generated by the methods and uses for the differentiated alveolar organoids are provided. The methods include digesting long-term expanding lung organoids (LO) into single cells (i.e., dissociated cells), and suspension culturing the dissociated cells in distal differentiation (DD) cell culture media in a non-adherent plate for an effective amount of time. Alveolar organoids includes a population of AT1 and AT2 cells and in suspension culture exhibit an apical-out polarity and include abundant cytoplasmic lamellar bodies and microvilli. The alveolar organoids can be utilized alone or in combination with 2D and 3D AWO, rebuilt the human respiratory epithelium in culture plates for studying biology and pathology of human respiratory system, including, but not limited to, the study of COVID-19 respiratory diseases.

Disclosed is a fiber-based hydrogel containing an aqueous hydrogel containing biocompatible microfibers having a high aspect ratio length to diameter suspended in water, the biocompatible microfibers mechanically interlocked. By simply adjusting the fiber aspect ratio, the fiber-based hydrogels can tune a considerable range of mechanical properties and drug release rates for clinical use. The fiber-based hydrogel has favorable biocompatibility and accelerated tissue regeneration ability demonstrated by a mice excision skin model.

A novel medical image generation approach synthesizes thin-cut computerized tomography (CT) images from thick-cut CT ones as inputs. First thick-cut CT images are obtained by maximizing the pixel-wise intensity of five or more than five continuous thin-cut ones after image registration. Second, the obtained thick-cut CT images are fed into a generator block which adopts an encoder-decoder architecture, where each thick-cut image is encoded into low-dimensional embedding space before decoding into multiple thin-cut ones. Third, a discriminator focuses on distinguishing original real thin-cut images from synthetic thin-cut images. An adversarial mechanism between the generator and discriminator causes the discriminator's output to provide an effective gradient update of the network parameters for the generator to increasingly improve the generator's ability to synthesize higher-quality thin-cut images and in turn promotes the discriminator's discriminating capability.

DEVICES AND METHODS FOR ORIENTING A SURGICAL DEVICE
Devices and methods are provided that simplify and improve placement of stabilizing orthopedic pins, wires, and/or screws in the pelvis in minimally invasive surgery. Provided is a surgical instrument guide containing an adjustable friction clamp for positioning and securing a cannula, and a probe with a blunt, nonpenetrating tip. The probe and a surgical instrument held by the friction clamp are oriented at right angles to one another. In use, the blunt probe is introduced through a small incision and advanced by blunt dissection until the blunt tip is positioned at a desired anatomical landmark on the surface of the pelvis. A cannula inserted through the friction clamp is advanced until the tip of the cannula is within 2 cm of the blunt tip of the probe; an orthopedic pin, wire, and/or screw can then be inserted through the cannula to provide stabilization. The blunt probe may be cannulated.

Abstract
Broad spectrum antiviral peptides and composition including therapeutically effective amounts of the antiviral peptides along with a pharmaceutically acceptable carrier are provided. The antiviral compositions show a strong broad spectrum antiviral effect, without resulting in viral resistance. Preferably, the antiviral compositions contain P16 or a P16-like peptide which can both inhibit viral fusion and endosomal acidification to prevent viral entry through the endocytic pathway.
