Abstract

Disclosed are methods for constructing disulfide mimetics for late-stage cyclization of peptides. The disclosed method provides late-stage cyclization via thioacetalization of peptides containing two cysteine residues to form thioacetal group. These bonds are stable under acidic, basic, and reduced conditions. The reaction can use a broad range of cyclic ketones even acetone as crosslinkers. Trifluoroacetic acid (TFA) alone can act as both the catalyst and the sole solvent, thus allowing most peptide sequences to be cyclized via the disclosed method. The disclosed method provides a number of benefits and advantages, such as the use of TFA as both a robust catalyst of the reaction and the sole solvent needed, the easy availability of acetone and of diverse cyclic ketones for use as the reactant, the good chemoselectivity and tolerance of native peptide to the reaction, and the fact that the resulting crosslinks are structural mimetics of disulfides.

Abstract

A steerable arm for use in endoscopic surgical procedures. The steerable arm cut from a tube of nitinol to provide flexibility and resilience by the structure into which the tube is cut. Typically, the cut tube comprises a spiralling coil, the loops of which are in contact on one lateral side of the tube and the tube is bent to open apart the loops on the other lateral side. The material which the tube is made of is capable of retaining memory of the bend. This provides a bias for restoring the tube to the bend whenever a force causing the tube to flex is removed.

Abstract

The subject invention pertains to a new method for measuring the elastic properties of microfibers by rope-coiling. Rope-coiling refers to the buckling of a slender elastic fiber caused by axial compression. A continuous flow microfluidic method enables the high-throughput measurement of the elasticity of microfibers by rope-coiling, where sample loading and unloading are not needed between consecutive measurements. In certain embodiments the coiling radius can be directly proportional to the elastic modulus of the fiber, facilitating calibration to measure fiber elasticity for high-throughput applications. Throughput can be thousands of times higher than that of a tensile tester, making possible an in situ, on-line measurement in a microfluidic production line, which couples the making of microfibers and the measurement of elasticity on the same line. The new method can also measure certain fibers with local variations in elasticity.

Abstract

A novel medical image generation approach synthesizes thin-cut computerized tomography (CT) images from thick-cut CT ones as inputs. First thick-cut CT images are obtained by maximizing the pixel-wise intensity of five or more than five continuous thin-cut ones after image registration. Second, the obtained thick-cut CT images are fed into a generator block which adopts an encoder-decoder architecture, where each thick-cut image is encoded into low-dimensional embedding space before decoding into multiple thin-cut ones. Third, a discriminator focuses on distinguishing original real thin-cut images from synthetic thin-cut images. An adversarial mechanism between the generator and discriminator causes the discriminator's output to provide an effective gradient update of the network parameters for the generator to increasingly improve the generator's ability to synthesize higher-quality thin-cut images and in turn promotes the discriminator's discriminating capability.

Abstract

Described herein are compounds containing palladium (II), as the central metal atom, and tetradentate [N^C^C^N] ligands. The compounds are charge neutral, and feature a donor-acceptor structure where a pendant substituted amino group (such as unsubstituted diphenylamine or substituted diphenylamine) and a heteroaryl group (such as pyridine group) serve as donor and acceptor, respectively. This donor-acceptor structure introduces a set of low-energy singlet and triplet charge-transfer excited states with small energy separation allowing for efficient thermally activated delayed fluorescence to take place.

Abstract

The NS1 protein of influenza virus is a key virulent element with multi-functional roles in virus replication and acts as a strong interferon (IFN) antagonist. A live attenuated virus (LAIV) is provided using a master backbone, which contains the influenza B (HK8038) virus and includes a deletion of the NSI coding region (DelNS1). The LAIV is based on novel adaptive mutations, which support DelNS1 influenza B live attenuated virus (LAIV) replication in vaccine producing cells. DelNS1 influenza B LAIV shows spontaneous cold adaption with preference to grow at 30-33° C. but restriction at 37-39° C. The LAIV can be used to protect a subject, against a lethal challenge of antigenic distant influenza B viruses. DelNS1 LAIV with adaptive mutations for growing in vaccine producing systems is an important strategy for making highly attenuated and immunogenic live attenuated influenza vaccines with the ability to induce broad cross protective immunity for seasonal influenza.

Abstract

Provided herein are novel ortho-Phthalaldehyde (OPA) containing linkers (OPA-L) and the use of OPA-L for the preparation of Antibody-drug conjugate (ADC) via the formation of Phthalimidine through the reaction of primary amine on antibody (e.g., residue of Lysine) and ortho-Phthalaldehyde. The advantage of this OPA-L is high reactivity and can be applied in different types of antibodies to form stably-linked conjugates. The use of OPA-L for the preparation of ADC is advantageous for mild and wide condition of conjugation, for instance, low percentage of organic solvent required, wide range of pH and temperature can be used.

Abstract

Systems and method for versatile multiplexed spinning/rotating bioassays are provided. This bioassay platform can take the advantage of the high-speed spinning motion, which naturally provides on-the-fly cellular imaging at the rate that cannot be reached by the conventional cameras or laser-scanning techniques, but ultrafast imaging modalities. More importantly, the functionalized solid substrates derived from the disk substrate can be compatible with adherent cell culture as well as biochemically-specific cell-capture, which can now be assayed with ultrafast imaging modalities at an ultra-high-speed line-scan rate of >10 MHz. Large-format spinning high-throughput imaging assay could thus be a potent tool for scaling both the assay throughput as well as content/multiplexity as demanded in many applications, e.g. drug discovery, and rare cancer cell screening.

Abstract

This invention is related to a dimming method and a dimming circuit for light-emitting-diode (LED) assemblies driven by nominally non-dimmable passive LED drivers. An LED system usually consists of an LED driver and an LED assembly. The LED driver normally provides a current source for the LED assembly. The LED assembly consists of individual LED devices or several LED modules mounted on a heatsink. LED devices or modules can be connected in series or in parallel or a combination of both. This invention involves the combined use of a controller and circuit for providing dimming function in the LED assembly driven by a passive LED driver without closed-loop current regulation. By using one or more controllable bypass-switches across sections of the LED strings in an LED assembly, the power of one or more groups of LED devices or LED modules can be cut off in order to dim the LED system. When the voltage of LED is reduced by closing the bypass switch or switches, a uniqueness of this invention is that the per-unit increase in the output current of the passive driver must be smaller than the per-unit voltage reduction of the LED load, resulting in reduction of power consumption in the LED load for dimming purpose. The dimming control can take turns to cut off the power of different groups of LED devices or modules in different times in order to share operating burden and preserve lifetime of the LED assembly. The invention is particularly suitable for, but not restrict to, lighting applications in smart lampposts in which smart wireless communication systems are mounted on the same lampposts as the LED assemblies and dimming signals can be received wirelessly. The advantages of this invention are that the dimming function is provided in the LED assembly rather than in the LED drivers, and the dimming circuit can be linked to smart control in emerging smart lampposts.

Abstract

AM580 and structurally related compounds have been found to be useful in treating infection by a wide range of RNA and DNA viruses, and also in reducing associated inflammation. This activity is independent of RAR-α signaling, and is not a result of activation of the hosts innate immune response. Broad antiviral activity of AM580 and structurally related compounds is a due to modulation of lipogenesis so as to correct disregulation of this pathway in virus-infected cells, via inhibition of nSREPBP.
