Abstract

The present invention relates to a composition for predicting cetuximab responsiveness in cancer, and more specifically to a composition for predicting cetuximab responsiveness, comprising a complex (Cetux-probe) having a fluorophore and a quencher bound to both sides of a peptide cleaved by a lysosomal enzyme, and having cetuximab conjugated to the C-terminus of the peptide. The Cetux-probe of the present invention operates in the same manner as the therapeutic mechanism of cetuximab, and thus the therapeutic effect and responsiveness of cetuximab may be quickly and accurately predicted. Cetux-probe activated fluorescence may be used to measure EGFR degradation, and a strong linear correlation between the Cetux-probe activated fluorescence and the cytotoxicity of cetuximab was observed in colorectal cancer cells and tumor-bearing mice. In addition, the predictive ability of the Cetux-probe activated fluorescence was confirmed to be much higher than that of EGFR expression or KRAS mutation status, and thus the Cetux-probe of the present invention may be usefully employed for predicting a response to cetuximab therapy by assessing EGFR degradation.

Abstract

The present invention relates to a compound sensing clathrin-mediated endocytosis, a fluorescent probe comprising same, and a sensor and a sensing method. The compound according to the present invention forms a micelle in a quenching state, and when in contact with a cell, the compound is transported into the cell through clathrin-mediated endocytosis and becomes fluorescent. Accordingly, the compound can be used to sense clathrin-mediated endocytosis or stain and label cells in which clathrin-mediated endocytosis takes place. The compound used in the present invention is particularly excellent in that the compound is stably transported via endocytosis into cells, such as live neurons or embryonic stem cells, within a relatively short time and maintains fluorescence for a long time at high fluorescence intensity. In addition, not only somatic cells but also cell-membranes and spikes can be stained.

Abstract

The present invention provides a linker having a net or cage structure, the structure of which is modified by a biomarker operating under a biphasic system consisting of a water layer and an oil layer. The linker is structurally modified in the presence of a biomarker to release a phase-shift probe contained therein, and the released phase-shift probe can be detected in the oil layer. In detection of the biomarker under the biphasic system using the linker of the present invention, the recognition of a target and the detection of a signal by the recognition are separated into the water layer and the oil layer, and thus the target can be detected with high accuracy. In addition, the system of the present invention does not require pretreatment of a sample, and thus can rapidly and accurately detect a quantitative biomarker with high sensitivity without expensive equipment and expertise required for pretreatment of a clinical specimen.

Abstract

Provided is BRCA-specific siRNA for enhancing the sensitivity of cancer cells to PARP inhibitors. According to the present disclosure, the siRNA can induce cancer cell death together with the PARP inhibitors in patients with wild-type BRCA genes with a low therapeutic effect of the PARP inhibitors, and the fusion protein-siRNA complex for siRNA delivery is up-taken into cells via CD47, and thus more specifically delivered to cancer cells while up-taken with high efficiency, thereby maximizing a desired cell death effect.

Abstract

The present invention provides a BRCA-specific siRNA for the purpose of improving the sensitivity of cancer cells to a PARP inhibitor. The siRNA of the present invention can induce cancer cell death together with a PARP inhibitor in patients having a wild-type BRCA gene with low therapeutic efficacy of the PARP inhibitor. A fusion protein-siRNA complex for siRNA delivery, of the present invention, is introduced into cells via CD47, and thus can be delivered more specifically to cancer cells while being introduced with high efficiency, thereby maximizing a desired cell death effect.

Abstract

There is provided a method and an apparatus for diagnosing an object placement error by using an artificial neural network by which feature data of the (N+1)-th stage among the feature data of a plurality of stages is generated by using any one feature data among a plurality of feature data generated in an operation process of a main network and feature data of the plurality of stages, and a placement error of at least one object is diagnosed by using an artificial neural network of a new structure, and thus, the diagnosis accuracy of a placement error of an object that is difficult to detect an edge, such as a transparent vial, may be greatly increased.
