Abstract

A method for feeding larval fish in an artificial habitat having a water level, includes the step of providing in the habitat a plurality of light sources at a plurality of three dimensional locations in the habitat and below the water level. Inanimate food particles are placed into the water. The plurality of lights are pulsed at the plurality of three dimensional locations intermittently to illuminate the food particles from a variety of different angles within the habitat. Secure data processing is described. Particular systems and methods involve enrollment units and methods, where the method includes obtaining an input data representing a raw data associated with a user, generating a template for the input data, and storing the template in an enrollment database, optionally with an identifier for the user. Other systems and method involve comparison or authentication units or methods, where the method involves obtaining templates corresponding to data sets to be compared, comparing the templates using a pre-defined comparison function to yield a similarity measure, and if the similarity measure meets a similarity criterion, determining that the data sets are from the same source. In the systems and methods, the templates are secure and noise tolerant templates configured to reveal limited features of the data set and to prevent reconstruction of the data set from the template.

Artificial (e.g., synthetic) carbohydrate receptors that specifically bind sialic acid and cancer cell surface carbohydrate antigens containing terminal sialic moieties (e.g., Lewis oligosacharide antigens) and methods of use were developed. The artificial carbohydrate receptors and methods can be used to detect and identify biologically important carbohydrates, selectively label cancer cells, to modulate cancer cell growth, and to modulate inflammation. Because the artificial carbohydrate receptors demonstrate a strong affinity for sialylated Lewis oligosacharide antigens (e.g., sialyl Lewis x (sLex) and sialyl Lewis a (sLea)) which are overexpressed on tumor cells during the onset and progression of cancer, and are overexpressed on the endothelium of chronically inflamed tissues, the artificial carbohydrate receptors described herein can find use as cancer diagnostic and therapeutic agents.

Abstract

A method of measuring the susceptibility of a subject to calcifying nanoparticle formation is disclosed. The method can include obtaining a cell culture comprising peripheral blood mononuclear cells, immune cells, or both, from a peripheral blood mononuclear fraction of a subject in a complete culture media; extracting a first aliquot and a second aliquot from the cell culture; and dispensing the first and second aliquot in different wells. The surface of the well containing the first aliquot can include a calcifying nanoparticle biofilm, while the well containing the second aliquot does not. The first and second aliquots can be incubated and the concentration of osteopontin in each aliquot determined. An osteopontin factor can be calculated and compared to osteopontin factors from a representative sample of other subjects. The osteopontin factor can be defined as the osteopontin concentration in the first aliquot divided by the osteopontin concentration in the second aliquot.

A method of characterizing calcifying nanoparticles (CNPs) can include creating a test sample comprising CNPs isolated from a biological source, a buffer solution, a plurality of calibration beads, and a fluorescent marker specifically linked to the CNPs; evaluating the test sample using a flow cytometer; and analyzing results from the flow cytometer to determine a characterizing feature of the calcifying nanoparticles. The characterizing feature of the calcifying nanoparticles can be the number of CNPs, concentration of CNPs, size of CNPs, level of CNP aggregation, size and light dispersion characteristics of CNPs, fluorescence intensity of the CNPs when labeled with a specific antibody, or a combinations thereof. The method can also include evaluating an isotype control comprising CNPs isolated from the biological source, the buffer solution, a plurality of calibration beads, and a fluorescent marker that is not linked to the CNPs.

Abstract

A new family Here we report a new family of four-cystine/three-loop conotoxins (designated framework 14). Three peptides of this family (flfl4a-c) were isolated from the venom of Conus floridanus floridensis and one (vill4a) from the venom of Conus villepinii, two worm-hunting Western Atlantic cone snail species. CD spectra and nanoNMR spectroscopy of these conotoxins directly isolated from the cone snails revealed a highly helical secondary structure for the four conotoxins. Sequence-specific nanoNMR analysis at room temperature revealed a well-defined helix-loop-helix tertiary structure that resembles that of the Cs &agr;/&agr; scorpion toxins κ-hefutoxin, K-KTx1.3 and Om-toxins, which adopt a stable three-dimensional fold where the two &agr;-helices are linked by the two disulfide bridges. One of these conotoxins (vill4a) has a Lys/Tyr (or Phe) diad, separated by approximately 6Å, which is a conserved structural feature in K+ channel blockers.
