Abstract

A microfluidic assembly including at least one microfluidic device is provided herein. The microfluidic assembly can be applied for vascularizing multicellular aggregates such as 2D or 3D animal cells or tissue implanted in a matrix that is held within an aperture of the microfluidic device. The microfluidic device in the microfluidic assembly is connected to a primary circulation system, which allows establishment and maintenance of physiologically relevant pressure gradients across the aperture of the microfluidic device. The microfluidic apparatus allows for connection of living microvessels, such as human microvessels, to a pressurized microfluidic channel via a porous membrane, allowing support of multicellular aggregates for long term culture.

Abstract

The present disclosure relates to a MEA electrolyser comprising a cathodic compartment operating CO2 reduction reactions (CO2RR) of CO2 from a gaseous CO2-containing stream, an anodic compartment operating all-liquid organic oxidation reactions (OOR), an ionic exchange membrane in between. A CO2RR-OOR system can further include a gas-liquid separation unit in fluid communication with the anodic compartment to receive the anodic product mixture and separate gaseous CO2 from the anodic product mixture to produce a CO2-depleted liquid product stream and a recovered pure gaseous CO2 stream. The system can further include a recycle line in fluid communication with the gas-liquid separation unit to redirect the recovered pure gaseous CO2 stream to the cathodic compartment of the MEA electrolyser as a portion of the gaseous CO2-containing stream. The present disclosure also concerns a process for electrochemically converting the gaseous CO2-containing stream to multi-carbon products in such a MEA CO2RR-OOR electrolyser.

Abstract

Described in this document is a process for making heat transfer devices such as cold plates. The cold plates can be made by 3-D printing a polymer substrate, applying thermal spraying deposition of metal over the substrate, dissolving the substrate for removal to form an enclosed flow region through for passage of heat transfer fluid. Molten metal droplets can be sprayed onto the substrate having a surface region comprising a water-soluble thermoplastic to form metal splats on the surface region at a splat temperature and surface properties such that the metal splats penetrate and interlock to form a solid metal coating adhered to the polymer substrate. A dissolvable substrate can facilitate removal by contacting with a solvent to form the enclosed flow region defined by metallic surfaces, and also can enable complex geometries and enhanced heat transfer performance without the need for extensive machining.

Methods and devices for identifying a pulmonary function pattern for a subject. A time series of measurements is obtained using an oscillometer. A fluidic wound dressing device comprises a branching network of fluidic channels connecting a single inlet port to a plurality of outlet ports in a biocompatible, treatment site-facing surface. The branching network enables the delivery of injected fluids containing therapeutic agents such as cells, drugs, antimicrobics, saline, and the like to the treatment site vis the outlet ports with a substantially uniform distribution. The outlet ports may be arranged in a regular grid array or other arrangement of substantially consistent density. The branching network may be based on a fractal arrangement such as an H-network to provide channels of substantially equal length connecting the inlet with each outlet. The relative cross-sectional areas of the channels may be determined in accordance with Murray's law to provide for optimal fluid delivery. The device may be used to deposit hydrogels comprising therapeutic cells such as fibroblasts and keratinocytes in layered structures.

The present disclosure provides a method of fabrication of physically crosslinked polyimide (PCPI) aerogels with significantly improved properties using no chemical crosslinker. The fabricated PCPI aerogels present high moisture resistance, hydrophobic behavior, ultralow density, ultrahigh porosity, excellent thermal stability, increased mechanical strength, and high mechanical flexibility. Also, samples have been successfully fabricated in a thin-film geometry with high mechanical flexibility and controlled thickness. Multilayer cathodes for the electrochemical reduction of carbon dioxide as well as membrane electrode assemblies and bipolar membrane electrode assemblies comprising the multilayer cathodes are described. Abstract

Novel chimeric peptides comprise an N-cadherin binding domain for binding N-cadherin, which is expressed on the surface of cells, in particular smooth muscle cells, and a fibronectin-binding domain, which binds surrounding extracellular matrix and a binding site for fibronectin. The chimeric peptides can be loaded on to nanoparticles, suitably degradable polar hydrophobic ionic polyurethane (D-PHI) nanoparticles for delivery, which can be incorporated into coatings for medical devices, including stents and balloons.

A system comprising a self-decarbonizing combustor and method of using the same are provided. The system comprises a combustor defining: a combustion chamber for combusting oxidizing agent and processed hydrocarbon. The combustor comprises an exhaust and a pyrolytic reactor which comprises a heating region and a separator. The heating region is defined within the combustion chamber for thermally coupling the heating region and the combustion chamber such that thermal energy generated in the combustion chamber is absorbed by the heating region to thermally decompose hydrocarbon feed. The separator is in fluid communication with the heating region for receiving hydrocarbon, where the separator is configured to pyrolyze the hydrocarbon and separate it into processed hydrocarbon and carbon black. The processed hydrocarbon is circulated to the combustion chamber for combustion and to heat the pyrolytic reactor. Carbon black may be removed from the separator as a separate product.

Abstract

Aptamers as nucleic acid affinity reagents are gaining traction in analytical applications owing to their high stability, compact size, and amenability to chemical modifications. It is highly desired to generate aptamers with different binding affinities to meet the requirements of various applications. However, systematic evolution of ligands by exponential enrichment (SELEX), the gold standard for aptamer generation, is unable to quantitatively produce aptamers with desired binding affinities and requires multiple rounds of selection to eliminate false positive hits. Here we introduce Pro-SELEX, a new approach for rapid discovery of aptamers with precisely defined binding affinities that combines efficient particle display, high-performance microfluidic sorting and high-content bioinformatics. Using the Pro-SELEX workflow, we were able to investigate the binding performance of individual aptamer candidates under different selective pressure in a single round of selection. Using human myeloperoxidase (MPO) as a target, we demonstrated that aptamers with dissociation constants spanning a twenty-fold range of affinities can be identified within one round of Pro-SELEX. Our selection strategy will have a significant impact on the fields of biosensing and bioseparation by generating aptamers with tailored binding affinities.

Various embodiments for AI-based systems and methods for engineering ionizable lipids for lipid nanoparticle (LNP)-based cargo molecule delivery are disclosed herein. In one example embodiment, the method comprises: designing a plurality of ionizable lipid structures based on a combination of a plurality of lipid headgroups, a plurality of first tailgroups and a plurality of second tailgroups; training a machine learning model based on the plurality of ionizable lipid structures; subsequently, selecting a first subset of ionizable lipid structures from the plurality of ionizable lipid structures and designating the first subset of ionizable lipid structures as experimental ionizable lipid structures; synthesizing, for example using a high throughput synthetic method, each of the experimental ionizable lipid structures; preparing a first set of separate LNPs, each LNP comprising a unique experimental ionizable lipid structure and the cargo molecule; determining a corresponding cargo molecule transfection efficiency value of each of the prepared LNPs in a target cell; further training the machine learning model based on the prepared LNPs and the corresponding cargo molecule transfection efficiency values; and deploying the further trained machine learning model to identify at least one lead ionizable lipid structure.

Abstract

An integrated charging topology is introduced for drivetrain solutions operating split-phase machines with variations are proposed both for single phase and multi phase variants. An active power decoupling variant is introduced using a single-input space vector decomposition and the usage of a power ripple phasor. This variant approach enables closed-loop control of the harmonic output power ripple (e.g., second harmonic), compensating for unmeasured effects, such as the interfacing inductance periodic energy storage, and allowing for a near-total reduction in the experimental setting. In operation, the control approach can include controlling the average power transfer between the grid and the battery and/or attenuating a second harmonic power ripple.

Abstract

There is provided systems and methods for generating formulations with improved performance and lower resource consumption. The population of formulations may be created by a Differential Evolution (DE) process. In each successive generation, lower performing formulations may be replaced with new formulations predicted to have better performance by a modeling pipeline. The modeling pipeline may perform a search for ML architectures and hyperparameter optimization for a suitable ML architecture, and then train one or more machine learning models to minimize error (otherwise maximizing the resulting score for a formulation). The machine learning models may be an ensemble of Random Forest models. These new formulations may form part of the next generation in the DE process. This modified version of the Differential Evolution process using machine learning techniques may result in populations of formulations which are superior in performance, and/or require fewer iterations of generations to achieve formulations which meet performance objectives.

A method to obtain a cache miss ratio curve where a memory blocks of a cache have variable block sizes. By stacking sets of counters, each set being for a different block size, a stack distance for variable block sizes can be obtained and used to determine a miss ratio curve. Such curve can then be used to select a cache size that is appropriate for an application without requiring excessive memory. Methods can be used for batches of request, can apply limits to block sizes, and rounding for intermediary block sizes, they can be used with pruning, and their space complexity can be held constant.

Methods and systems for mRNA analysis and quantification of mRNA expression in cells are provided. An example method includes introducing a first capture probe and a second capture probe into the cells, the first capture probe and the second capture probe each configured to be complementary to a respective section of target mRNA within the cells, wherein binding of the first and second capture probes to the respective sections of the target mRNA results in tagging of the cells and causes the first and second capture probes to form clusters with each other. The first capture probe and the second capture probe are each bound to magnetic nanoparticles (MNPs) that, when trapped within the tagged cells, cause the tagged cells to be susceptible to magnetic forces.
