Described in this document is a process for making heat transfer devices such as cold plates. The cold plates can be made by 3-D printing a polymer substrate, applying thermal spraying deposition of metal over the substrate, dissolving the substrate for removal to form an enclosed flow region through for passage of heat transfer fluid. Molten metal droplets can be sprayed onto the substrate having a surface region comprising a water-soluble thermoplastic to form metal splats on the surface region at a splat temperature and surface properties such that the metal splats penetrate and interlock to form a solid metal coating adhered to the polymer substrate. A dissolvable substrate can facilitate removal by contacting with a solvent to form the enclosed flow region defined by metallic surfaces, and also can enable complex geometries and enhanced heat transfer performance without the need for extensive machining.

Aptamers as nucleic acid affinity reagents are gaining traction in analytical applications owing to their high stability, compact size, and amenability to chemical modifications. It is highly desired to generate aptamers with different binding affinities to meet the requirements of various applications. However, systematic evolution of ligands by exponential enrichment (SELEX), the gold standard for aptamer generation, is unable to quantitatively produce aptamers with desired binding affinities and requires multiple rounds of selection to eliminate false positive hits. Here we introduce Pro-SELEX, a new approach for rapid discovery of aptamers with precisely defined binding affinities that combines efficient particle display, high-performance microfluidic sorting and high-content bioinformatics. Using the Pro-SELEX workflow, we were able to investigate the binding performance of individual aptamer candidates under different selective pressure in a single round of selection. Using human myeloperoxidase (MPO) as a target, we demonstrated that aptamers with dissociation constants spanning a twenty-fold range of affinities can be identified within one round of Pro-SELEX. Our selection strategy will have a significant impact on the fields of biosensing and bioseparation by generating aptamers with tailored binding affinities.

There is provided systems and methods for generating formulations with improved performance and lower resource consumption. The population of formulations may be created by a Differential Evolution (DE) process. In each successive generation, lower performing formulations may be replaced with new formulations predicted to have better performance by a modeling pipeline. The modeling pipeline may perform a search for ML architectures and hyperparameter optimization for a suitable ML architecture, and then train one or more machine learning models to minimize error (otherwise maximizing the resulting score for a formulation). The machine learning models may be an ensemble of Random Forest models. These new formulations may form part of the next generation in the DE process. This modified version of the Differential Evolution process using machine learning techniques may result in populations of formulations which are superior in performance, and/or require fewer iterations of generations to achieve formulations which meet performance objectives.
