Abstract

The subject invention pertains to a long-term antigen depot that can bypass FBR and engage DCs to mature and migrate to lymph nodes to activate antigen-specific T-cell activations. Leveraging on the immunomodulatory properties of exogenous polysaccharides and the anti-fouling characteristics of zwitterionic phosphorylcholine (PC) polymer, the invention features a PC functionalized dextran (PCDX) hydrogel for long-term antigen delivery. PCDX in both injectable scaffold and microparticles (MPs) forms effectively evades FBR and provides slow release of antigens resulting in local enrichment of CD11c+ DCs at the MPs injection sites. DC cultured on PCDX exhibits strong immunogenic activation with high CD86, CD40, MHC-I/peptide complex. PCDX also generates DC with propensity in migration to lymph nodes, as well as antigen presentation to trigger both CD4+ and CD8+ arms of T-cell responses. Besides cellular responses, PCDX can also induce potent humoral responses, with high levels of antigen specific IgG1 and IgG2a by day 28. Overall, PCDX provides long-term delivery of antigens for vaccine development.

Abstract

A system using global and local-aware denoising framework for commonsense knowledge graph noise detection is provided. The system includes an input receiver, a triple encoder, a global noise detector, a local noise detector, and a comprehensive evaluation scorer. The triple encoder receives the CSKG information from the input receiver and extract semantic information from free-text formatted nodes in the CSKG information, so as to obtain structured triples. The global noise detector identifies high-frequency patterns through rule mining and uses a rule encoder to generalize learned rules and guide noise detection. The local noise detector utilizes the structured triples output by the triple encoder to aggregate neighboring triple information around both the head and tail nodes via graph networks. The comprehensive evaluation scorer provides a comprehensive score based on global noise detection and local noise detection, so as to indicate a noise level for the CSKG information.

Abstract

Provided herein is a method of improved commonsense knowledge bases reasoning through constraint-based prompting. A novel dual-module constraint-based prompt engineering system named ConstraintChecker is involved, which employs a first rule-based module to produce a list of constraints and a second zero-shot learning module to check whether the knowledge instance satisfies all constraints. The acquired constraint-checking result is then aggregated with the output of the main prompting backbone to produce the final output. This present method is more effective in commonsense knowledge bases reasoning then that by employing the prompting backbone alone, and also significantly reduces computational costs.

Abstract

Provided herein is a novel method named Knowledge-Constrained Tree Search Decoding (KCTS) is provided, which is a first discriminator-guided constrained decoding approach to knowledge-grounded generation. The KCTS method also employs a Monte-Carlo Tree Search decoding (MCTS) instead of conventional LM decoding methods, which re-orders each token's importance based on the estimated future faithfulness starting from that token with multiple simulations. Coupled with a novel token-level hallucination detection method, RIPA (Reward Inflection Point Approximation), trained to predict the starting point of the hallucinating token, enabling effective adaptation of the knowledge classifier defined on the sequence level to the token level, this novel method is able to greatly mitigate hallucination problems of LLMs without the need of fine-tuning, thereby preventing the incurrence of high computational costs.

Abstract

Single atom catalysts (SACs) are efficient electrocatalysts for catalyzing the oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) in lithium-oxygen batteries (LOBs). Our method is to study the origin of reactivity for a series of 17 transition metals supported on nitrogen-doped graphene, denoted SACs using Quantum Mechanics methods. Based on Gibbs free energy calculations, Zn-SAC performs the highest electrochemical activity among 17 SACs investigated. Machine learning (ML) is developed an intrinsic descriptor Φ, that correlates the catalytic activity with electronic and chemical properties of the catalytic centers at the M-N4 active site on graphene surface. A linear relationship between Φ and the catalytic activity that provides guidance for designing efficient SACs for electrocatalysis in LOBs. Zn-SAC is synthesized through chemical vapor deposition (CVD) process, which shows more stable cyclability and exhibits smaller reaction overpotentials, verifying the simulation results.

Abstract

A passive on-chip optical long-pass filter for removing residual pump photons at short wavelengths after nonlinear generation of photons. A thin layer (<100 nm) of amorphous or poly-crystalline silicon is deposited onto a section of a waveguide to absorb light with a wavelength shorter than the silicon bandgap wavelength of ˜1.1 μm, while the nonlinearly generated light in longer wavelengths than the silicon bandgap wavelength propagates in the waveguide with a negligible absorption loss. The filter is applicable to attain an on-chip optical pump light rejection ratio exceeding 120 dB for nonlinear and quantum photonic chips. The filter is conceptually simple to design and can be fabricated by a CMOS process with potentially a high wafer-level scalability and manufacturability at a low cost. The filter can be realized in various integrated photonic platforms, including silicon carbide, silicon nitride, lithium niobate and aluminum nitride.

Abstract

A machine learning (ML) workflow for virtual staining and computational pathology based on a label-free tissue sample is provided to avoid a laborious and time-consuming procedure of pathologist-based interpretation of formalin-fixed and paraffin-embedded histology. A virtual-staining ML model translates a label-free autofluorescence (AF) whole slide image (WSI) of the tissue sample into a virtual hematoxylin and eosin (H&E) -stained WSI, which is further transformed into a WSI virtually stained with a special stain by a stain-transformation ML model. Each of the ML models can be realized as a U-Frame model. A multiple instance learning (MIL) network with attention-based pooling can be applied to the virtual H&E-stained WSI or AF WSI to provide fast and accurate tumor diagnosis. Furthermore, the attention-based MIL network can be configured as a clustering-constrained-attention multiple instance learning (CLAM) model for providing multiclass tumor subtyping, or even multiclass disease typing, in diagnosis of the tissue sample.

Abstract

This invention refers to a liquid-based micro thermal convective accelerometer (MTCA) optimized using a compact model based on the Rayleigh number (Ra). The MTCA is fabricated using CMOS MEMS technology. To ensure water resistance, an isolation layer such as a waterproof cover, exemplified by the conformal Parylene C coating, is employed. The device's performance is assessed in terms of sensitivity, response time, and noise. Theoretical and experimental findings establish that fluids with higher Ra numbers yield improved MTCA performance. Ra-based model showed its advantage to make a more accurate prediction than the simple linear model to select suitable fluid to enhance the sensitivity and balance the linear range of the device. In some cases, the liquid of MTCA can be selected as alcohol, and an alcohol-based MTCA was achieved with a two-order-of magnitude increase in sensitivity and one-order-of-magnitude decrease in the limit of detection compared with the air-based MTCA.

Abstract

A heat spreader composed of a hexagonal boron carbon nitride (hBCN) thin film, can be a major solution for the ever-increasing problem of overheating electronic devices. This heat spreader can dissipate heat away from hotspots on electronic devices, unlocking additional processing power. Our method is able to effectively tune the thermal and electrical conductivity of our films by modulating the carbon content of the precursor used in a chemical vapor deposition (CVD) process. Using this method, we can generate a heat spreader that is electrically insulative, so it will not have the potential to create shorts, like graphene and metals. Additionally, we can maximize the thermal conductivity, obtaining a value of 460±149 W/m·K, which rivals or even surpasses hexagonal boron nitride (hBN). As our precursor material is 10 times cheaper than the most common hBN precursor, this is an economical alternative to hBN for heat spreader applications.

Abstract

A method and an apparatus for decoding channel codes received from a noisy communication channel are provided. The method comprises: demodulating the received channel codes to obtain a demodulated data vector; and decoding, using a compute-in-memory (CIM), the demodulated data vector to obtain one or more candidate output codewords, the demodulated data vector is decoded with a guessing random additive noise decoding (GRAND) algorithm to obtain a candidate output codeword. By leveraging CIM technique for GRAND implementation, due to the intrinsic small size of memory cells relative to logic gates, the integration of storing and computing operations (e.g. multiply-and-accumulate (MAC) operation on the input vector with the weight matrix) in memory cells enables concurrently evaluating a multitude error patterns, therefore substantially reduce the worst-case latency of GRAND hardware implementation.

Abstract

Translational rapid ultraviolet-excited sectioning tomography (TRUST) applies ultraviolet (UV) excitation to a sample and images fluorescence and autofluorescence emission for tomographically imaging the sample. Deep-learning neural networks are used to achieve higher imaging speed and imaging resolution. In one use, fluorescence images acquired with relatively low imaging resolution can be transformed into high-resolution images through the first conditional generative adversarial network (cGAN), a super-resolution neural network (e.g., ESRGAN), which is also helpful for reducing the image scanning time. In another use, the second cGAN, such as Pix2Pix, is used to realize virtual optical sectioning to enhance the axial resolution of the imaging system. Compared to the conventional pattern illumination methods (e.g., HiLo microscopy), which need at least two shots for each field of view, the imaging speed is also times higher because only one shot under the uniform-illumination condition of UV irradiation is required.

Abstract

Microfluidic devices having microwell arrays that enable scalable and parallel droplet multi-step reactions and associated methods for implementing reactions in paired microwells of two different sizes via numerous repeating units of trapping wells as well as sequentially collecting and merging droplets. Droplets that contain the desired species or reagents for reactions are input onto the microfluidic platform. Each well is set up to initially capture one larger droplet, then repeatedly capture smaller ones, followed by merging of larger and smaller droplets. In some embodiments, this action is facilitated by tilting the chip. In some implementations, larger droplets merge three batches of paired, smaller droplets with various needed components in succession while conducting incubation in between the merge and loading. In alternative implementations, each trapping well has two or more active capture sites that merge multiplexed smaller droplets simultaneously and merge more than once.

Abstract

A technique for detection, classification and location identification of defects in a pressurized pipe network uses supervised machine learning and transient response signals (TRSs) of potential defect scenarios as training data. The TRSs are obtained from a model and/or measurements. The TRSs are arranged in an appropriate matrix form and its singular vectors (SVs) are obtained using singular value decomposition. The defect detection and classification procedure is conducted by projecting a measured TRS of the pipe network into a SV space. The location of the measured TRS in the SV space indicates the state of defective pipe section (if any), classify the defect according to clusters of training data (e.g., leak, blockage), and locate the defect within an identified defective section. The technique can accurately detect defective pipes in a network with a training data set as small as three scenarios per pipe section and one single measurement location.

Measured multi-input-multi-output (MIMO) imaging data at high frequency (10kH-100 kHz) are processed by an adapted Time Reversal-MUltiple SIgnal Classification (TR-MUSIC) algorithm to provide a high-resolution image of a pipeline section about 100 m long. The resultant image reveals the pipe wall inner and outer condition, and incipient and existing defects at a scale up to millimeters. The technique is tested and validated in a laboratory environment and in a large-scale facility on pressurized water-filled viscoelastic high-density polyethylene pipes. Furthermore, the technique can be applied to different fluids and pipe materials. In addition, the technique is computationally efficient, and nondestructive. The technique requires only local intrusion for placing transducers at an access point.

Abstract

A high-power single-frequency laser system and a method for implementing the same. The system and method require only a photodetector (110) and a narrow optical bandpass filter (108) to probe the spectral mode of slave laser (106), no additional bulky devices are required. The photodetector (110) and narrow optical bandpass filter (108) can be easily integrated into the beam path. The spectral mode of slave laser (106) is monitored through the reflected signal from the narrow-laser line filter (108), which is neither sensitive to beam alignment nor bandwidth limited. As a result, high spectral mode purity and low intensity noise of the slave laser (106) can be simultaneously obtained. The slave laser (106) can be locked even when the master laser (102) frequency is dynamically scanned over GHIz range. The system and method can be applied in laser cooling and trapping of atoms towards degenerate dipolar quantum gases.

Abstract

A rapid, label-free, non-destructive imaging method and system (100) for unprocessed biological tissue, which is based on microscopy with ultraviolet single plane illumination (MUSI). The present system (100) or method employs a selective two-axis planar illumination configuration, which decouples the illumination from the detection path, and utilize the intrinsic fluorescence of certain endogenous fluorophores from biological tissues as a natural source compared to deep UV illumination sources. In contrast to images produced by clinical standard methods (i.e. H&E staining of formalin-fixed and paraffin-embedded tissues), images generated by MUSI reveal comparable or even better diagnostic features, providing clinicians, pathologists, and surgeons with greater potential as post-operative and intra-operative diagnostic tools.

Abstract

The present invention discloses aggregation-induced emission fluorescent compounds with near-infrared (NIR) emission. This fluorescent compound is then encapsulated by polymer matrix to yield nanoaggregates which is confirmed to show an excellent photothermal conversion efficiency while maintaining a high fluorescence quantum yield. Consequently, the photothermal therapeutic efficacy can be improved by PTA-oriented accumulation of nanoaggregates, compared with the barely enhanced permeability and retention (EPR) effect. The in vitro and in vivo verification further confirmed that the nanoaggregates exhibit superior fluorescence-guided phototheranostics ability to eliminate tumors.
