Abstract

The present invention relates to exosomes comprising an miRNA pool based on the inhibition of TGF-β signaling involved in cancer growth and metastasis. The exosomes, which are derived from tripartite motif containing 40 (TRIM40) transformed cells treated with staurosporine, comprise a pool of miRNAs that inhibit the expression of ECM- and EMT-related genes, the miRNAs include miR-1343-3p (which inhibits the expression of TGBR1 and TGFBR2), miR-4285 (which inhibits the expression of the TGFB1 gene), and miR-194-3p (which inhibits the TGF-beta signaling pathway), which inhibit the TGF-beta signaling pathway in cancer cells, thereby comprehensively blocking the expression of EMT- and ECM-related genes by surpassing a one-dimensional method of loading one specific miRNA or a small molecule that exhibits anticancer activity so as to inhibit TGF-beta signaling pathways when the miRNAs are absorbed and delivered into cancer cells, and thus the cancer microenvironment is modulated such that strong anticancer responses can be induced in preclinical settings.

Abstract

One of the most important technical problems to be solved in the present specification is that of providing a prime editor capable of exhibiting an actual therapeutic effect by selecting a prime editor exhibiting an effect of correcting c.4957dupA mutation of an EYS gene of actual human retinal cells from among numerous theoretically designable prime editors and specifying a configuration thereof. In order to solve this technical problem, the present specification provides a prime editor and a component thereof, wherein the prime editor is screened through a high-throughput screening method, is verified in actual human retinal cells, and can actually correct the c.4957dupA mutation of the EYS gene. The prime editor disclosed in the present specification acts on human retinal cells including the c.4957dupA mutation in the EYS gene, thereby being able to correct the EYS gene to a normal state. Therefore, the prime editor can be used as a therapeutic agent for retinitis pigmentosa caused by the c.4957dupA mutation in the EYS gene.

Abstract

The present invention relates to an autoimmune disease animal model and a method for preparing the same. The animal model of the present invention efficiently generates an autoantibody that causes autoimmune disease, by introducing an autoantigen-encoding gene through the Cre-LoxP system, which enables stable and continuous expression of the autoantigen at a desired time by treatment of Cre recombinase. Contrary to the conventional method that fails to effectively produce autoantibody due to leaky expression and immune tolerance, or requires repeated administration over several weeks, the present invention achieves sustainable expression of an effective amount of autoantigen by only one TAT-Cre recombinase treatment, thus may be utilized as an outstanding animal model reproducing various symptoms and molecular mechanisms of autoimmune diseases.

Abstract

The present invention relates to a method for prevention, alleviation, or treatment of cancer patients, especially epithelial mesenchymal transition (EMP)-subtype cancer patients. If PHGDH, SHMT and MTHFD2 inhibitors are co-administered to patients with cancer in which the expression level of a glutaminase (GLS) gene or a protein encoded thereby has increased, 1C metabolism is more effectively inhibited such that there is a synergistic effect on the inhibition of cancer cell proliferation in patients with refractory cancer that is difficult to treat because of recurrence, metastasis, and anticancer drug resistance, thereby enabling cancer to be very effectively treated. In addition, the expression level of the GLS gene or protein encoded thereby is measured so that information related to customized treatment methods from initial stages is provided to each patient and the success of treatment can be increased.

Abstract

Provided herein are novel reprogrammed smooth muscle cells (rSMCs) and methods of making and using the cells for the treatment of ischemia. The rSMCs are produced by culturing a fibroblast with an all-trans-retinoic acid (ATRA) under conditions that produce the rSMC from the fibroblast, wherein the fibroblasts are genetically modified to overexpress myocardin. The rSMCs offer advantages over currently available regenerative vascular therapies by promoting vascular perfusion in a recipient subject. In particular, the rSMCs can increase neovascularization of both small and large vessels.

Abstract

Pancreatic ductal adenocarcinoma (PDAC) is the fourth leading cause of cancer-related deaths worldwide, but lacks effective prognostic indicators. Methionyl-tRNA synthetase 1 (MARS1), which is an important enzyme in translation initiation for delivering Met to initiator tRNA, is associated with cancer development and progression. MARS1 expression was found to significantly increase in PDAC compared to normal pancreatic duct tissue. In addition, the high expression of MARS1 was found to be associated with poor prognosis in PDAC patients. This suggests that MARS1 is involved in pancreatic carcinogenesis and has potential as a novel prognostic indicator for PDAC.

Abstract

The present invention relates to a method for predicting the response to immunotherapeutic agents, comprising a step of measuring, in cancer cells or cancer tissue, the expression level of a dexamethasone-induced ras-related protein 1 (Dexras 1) gene or a protein encoded thereby. According to the present invention, research has been carried for prediction of the response to immunotherapeutic agents and, particularly, research has been carried out for prediction of the response to immunotherapeutic agents, highly reliable therapeutic agents and effective diagnosis of stem cell-like gastric cancer, which has the poorest prognosis and for which discovery of an effective treatment and diagnosis is difficult. As a result, it has been identified that in refractory carcinoma, the expression of Dexras 1 is high and, when an immunotherapeutic agent is injected, resistance to the immunotherapeutic agent is exhibited. In addition, changes in the expression level of Dexras 1 in cancer samples are measured such that immunotherapeutic agents for refractory cancer, such as stem cell-like gastric cancer that is difficult to be treated, are provided and refractory cancer can be diagnosed, and the response to anticancer immunotherapeutic agents for effective immunotherapy can be predicted such that the present invention has been completed. Therefore, the present invention is expected to be widely used in the development of novel therapeutic agents for refractory cancer.
