Abstract

The present invention relates to substituted benzophenones of structural Formula I employed as small molecule inhibitor for controlling obesity. The present invention further relates to screening of small molecule inhibitors against p300 which selectively targets histone butyrylation thereby specifically inhibiting adipogenesis further aiding to prevention of weight gain. The present invention reveals that histone butyrylation increases during adipogenesis. Hence, inhibition of histone butyrylation by a small molecule inhibitor would be a promising therapeutic strategy to control obesity. Significantly, besides inhibiting adipogenesis in cellular model, it also prevents weight gain in high fat diet based mouse model system, indicating the possible use of the scaffold for obesity control.

The nanosphere is carbon nanosphere (CSP) which is intrinsically fluorescent and the HAT activator is N-(4-Chloro-3-trifluoromethyl-phenyl)-2-n-propoxy-benzamide. The N-(4-Chloro-3-trifluoromethyl-phenyl)-2-n-propoxy-benzamide is covalently conjugated with the carbon nanosphere. The present invention further relates to a process for obtaining a composition including carbon nanosphere and Histone acetyltransferase (HAT) activator [N-(4-Chloro-3-trifluoromethyl-phenyl)-2-n-propoxy-benzamide]. The composition is capable of crossing blood brain barrier and inducing histone acetylation in brain. Further, the composition is capable of increasing neurogenesis, as well as improving long-term memory formation. The composition manages pathological conditions to a subject in need thereof, such as aging-related, neurodegenerative diseases (Alzheimer's in particular), neurological disorders, depression or other kinds of diseases in which increased HAT activity, neurogenesis and/or memory improvement would benefit.

Abstract

The present disclosure is in relation to a composition comprising nanosphere and histone acetyltransferase (HAT) activator. The disclosed nanosphere is carbon nanosphere (CSP) which is intrinsically fluorescent and the HAT activator is N-(4-Chloro-3-trifluoromethyl- phenyl)-2-n-propoxy-benzamide [COMPOUND 1]. The said N-(4-Chloro-3-trifluoromethyl- phenyl)-2-n-propoxy-benzamide is covalently conjugated with the carbon nanosphere. The present disclosure further relates to a process for obtaining a composition comprising carbon nanosphere and Histone acetyltransferase (HAT) activator [N-(4-Chloro-3-trifluoromethyl- phenyl)-2-n-propoxy-benzamide]. The composition is capable of crossing blood brain barrier and inducing histone acetylation in brain. Further, the composition is capable of increasing neurogenesis, as well as improving long-term memory formation. The said composition thus provides for managing pathological conditions to a subject in need thereof, such as aging- related, neurodegenerative diseases (Alzheimer's in particular), neurological disorders, depression or other kinds of diseases in which increased HAT activity, neurogenesis and/or memory improvement would benefit.

Abstract

Embodiments of the present disclosure relate to a system and method to detect hydrogen leakage. The system uses a fluid sensing apparatus (104), a light source (120) and and photo detector (122). The nano-crystallised palladium gratings (118) are used as sensors which expand sensitively upon exposure to the hydrogen ( H2). In an embodiment, the hydrogen sensing is based on monitoring the changes in the diffraction efficiency (DE) which is defined as the ratio of the first and the zeroth order diffracted beam intensities. The diffraction efficiency undergoes large and sudden changes as the nano-crystalline Pd grating becomes highly disordered due to PdHx formation. An embodiment of the present disclosure also relates to producing nanocrystalline Pd diffraction gratings along with the design and fabrication aspects of an indigenously built optical diffraction cell for H2sensing.
