In this technology, we developed AAP, an ALDH1A1-activated photosensitizer that leverages d-PeT suppression to regulate intersystem crossing and singlet oxygen generation. Through systematic tuning of halogen substitution and oxidation-dependent activation, we demonstrated that mono-iodination at the xanthene core provides an optimal balance between ISC enhancement and background suppression, enabling activation-dependent phototoxicity. In vitro and in vivo studies confirmed that AAP selectively targets ALDH1A1-expressing cells, with minimal off-target effects and strong photodynamic efficacy upon irradiation.

In this work, we have developed a luciferin derivative, AlDeLuc, the first logic-gated bioluminescence probe for cancer stem cells (CSCs) that is sequentially activated by acidic environments and ALDH1A1 activity. Beyond demonstrating efficacy in multiple cancer cell lines and a murine model of breast cancer, we employed AlDeLuc to investigate how the CSC population is altered by the inflammatory status of a tumor through a high-fat diet. The expression of the P-glycoprotein (P-gp) efflux transporter at the blood-brain interface impedes BBB penetrance of most small molecules.
