The present technology provides a genetically modified immune cell expressing a chimeric antigen receptor (CAR) and also expressing an elevated level of a ligand for a tumor necrosis factor (TNF) receptor. The genetically modified cell takes advantage of dual pathways for killing cancer cells, cellular pathogens, and cells infected with viruses. The CAR- mediated pathway promotes killing action by perforin and granzymes released by cytotoxic T lymphocytes and natural killer cells, as well as the TNF receptor (i.e., death domain) mediated target cell killing by these same cells. Provided herein are methods and systems for a hard drive failure prediction system to determine a predicted hard drive failure within a time-in-advance determined time period. The system includes a machine-learning model, such as an XGBoost model, trained using historical hard drive operational data from a data center. The historical hard drive operation data corresponds to SMART attributes. Statistical analysis is performed to determine a set of SMART attributes most indicative of hard drive failure. The model is trained using the historical hard drive operational data corresponding to the set of SMART attributes. The accuracy results of the model are used to determine a time-in-advance value that balances accuracy with failure lead time. A signal length of operational data for a particular hard drive is provided as input to the trained model and an output indicates if the hard drive will fail within the time period of the time-in-advance value.

Voltage-gated sodium (NaV) channels are densely expressed in most excitable cells and activate in response to depolarization, causing a rapid influx of Na+ ions that initiates the action potential. The voltage-dependent activation of NaV channels is followed almost instantaneously by fast inactivation, setting the refractory period of excitable tissues. The gating cycle of NaV channels is subject to tight regulation, with perturbations leading to a range of pathophysiological states. The gating properties of most ion channels are regulated by the membrane phospholipid, phosphatidylinositol (4,5) bisphosphate (PI(4,5)P2). However, it is not known whether PI(4,5)P2 modulates the activity of NaV channels. Here, we utilize optogenetics to activate specific, membrane-associated phosphoinositide (PI)-phosphatases that dephosphorylate PI(4,5)P2 while simultaneously recording NaV1.4 channel currents. We show that dephosphorylating PI(4,5)P2 left-shifts the voltage-dependent gating of NaV1.4 to more hyperpolarized membrane potentials, augments the late current that persists after fast inactivation, and speeds the rate at which channels recover from fast inactivation. These effects are opposed by exogenous diC8PI(4,5)P2. We provide evidence that PI(4,5)P2 is a negative regulator that tunes the gating behavior of NaV1.4 channels.

Reduction in the tortuosity of electrodes is a favored strategy to enhance the fast-charging capability of lithium-ion batteries by optimizing the ion-transfer kinetics. A facile, low-cost, highly controllable, and high-output continual additive manufacturing roll-to-roll screen printing technology is disclosed herein to render customized vertical channels within electrodes. High-accuracy vertical channels were fabricated by applying as-developed inks, using LiNi0.6Mn0.2Co0.2O2, for example, as the cathode material. The optimized screen-printed electrode exhibited a seven-fold higher specific charge capacity (72 mAh/g) at a current rate of 6 C and superior stability compared with that of the conventional bar-coated electrode (10 mAh/g, 6 C) at a mass loading of 10 mg/cm2.

Hybrid structured materials are composed of hard cell structures connected by soft components such as soft networks, soft hinges, or bilayer joints. The soft components are responsive to external stimuli such as mechanical loads, temperature changes, humidity, and electric-magnetic fields.
